Resting state functional connectivity (RSFC) has been widely studied in functional magnetic resonance imaging (fMRI) and is observed by a significant temporal correlation of spontaneous low-frequency signal fluctuations (SLFs) both within and across hemispheres during rest. Different hypotheses of RSFC include the biophysical origin hypothesis and cognitive origin hypothesis, which show that the role of SLFs and RSFC is still not completely understood. Furthermore, RSFC and age studies have shown an "age-related compensation" phenomenon. RSFC data analysis methods include time domain analysis, seed-based correlation, regional homogeneity, and principal and independent component analyses. Despite advances in RSFC, the authors also discuss challenges and limitations, ranging from head motion to methodological limitations.

Highly abundant in neurons, the cellular prion protein (PrP) is an obligatory precursor to the disease-associated misfolded isoform denoted PrP that accumulates in the rare neurodegenerative disorders referred to either as transmissible spongiform encephalopathies (TSEs) or as prion diseases. The ability of PrP to serve as a substrate for this template-mediated conversion process depends on several criteria but importantly includes the presence or absence of certain endoproteolytic events performed at the cell surface or in acidic endolysosomal compartments. The major endoproteolytic events affecting PrP are referred to as α- and β-cleavages, and in this review we outline the sites within PrP at which the cleavages occur, the mechanisms potentially responsible and their relevance to pathology.

Mitochondria have generated the bulk of ATP to fuel cellular activities, including membrane trafficking, since the beginning of eukaryogenesis. How inhibition of mitochondrial energy production will affect the form and function of the endomembrane system and whether such changes are specific in today's cells remain unclear. During the trunk to tail transition the mammalian embryo builds the outlets for the intestinal and urogenital tracts, lays down the primordia for the hindlimb and external genitalia, and switches from the epiblast/primitive streak (PS) to the tail bud as the driver of axial extension. Genetic and molecular data indicate that Tgfbr1 is a key regulator of the trunk to tail transition. Tgfbr1 has been shown to control the switch of the neuromesodermal competent cells from the epiblast to the chordoneural hinge to generate the tail bud.
