Background: Huntington's disease (HD) is an autosomal dominant condition causing severe neurodegeneration in the striatum and the entorhinal cortex (EC). An epigenome wide association study of DNA methylation in HD by our group, identified potential hypomethylation at the PTGDS gene in the striatum. We aimed to validate this result through pyrosequencing, examining the locus in fine detail, and to assess the signal specificity by profiling multiple neurodegenerative diseases.

Background: In-vivo magnetic resonance imaging (MRI) has recently shown that patients with clinically diagnosed Alzheimer's disease (AD) and dementia with Lewy bodies (DLB) exhibit degeneration of the cholinergic nucleus basalis of Meynert and its white matter (WM) projections through the cingulum and external capsule pathways. Here, we propose an imaging-pathologic validation study aimed at investigating cholinergic WM pathways using post-mortem MRI of autopsy-confirmed AD, Lewy body dementia (LBD), and other neurodegenerative diseases (OTH).

Method: We included 53 brain donors (34 AD, 10 LBD, and 9 OTH, mainly including frontotemporal lobe degeneration and vascular disease, Table 1).

Background: Limbic-predominant age-related TDP-43 encephalopathy neuropathological change (LATE-NC) is a pathological process diagnosed at autopsy, involving deposition of TDP-43 in the medial temporal lobes. The name LATE-NC was recently proposed to represent the pathological process, while "LATE" has been suggested to represent the clinical syndrome. However, there are currently no available criteria to diagnose this syndrome during life, and the clinical phenotype is not well understood.
