Article Abstract

Natural killer (NK) cell repertoires are made up of phenotypically distinct subsets with different functional properties. The molecular programs involved in maintaining NK cell repertoire diversity under homeostatic conditions remain elusive. Here, we show that subset-specific NK cell proliferation kinetics correlate with mTOR activation, and global repertoire diversity is maintained through a high degree of intra-lineage subset plasticity during interleukin (IL)-15-driven homeostatic proliferation in vitro. Slowly cycling sorted KIRCD56 NK cells with an induced CD57 phenotype display increased functional potential associated with increased transcription of genes involved in adhesion and immune synapse formation. Rapidly cycling cells upregulate NKG2A, display a general loss of functionality, and a transcriptional signature associated with increased apoptosis/cellular stress, actin-remodeling, and nuclear factor κB (NF-κB) activation. These results shed light on the role of intra-lineage plasticity during NK cell homeostasis and suggest that the functional fate of the cell is tightly linked to the acquired phenotype and transcriptional reprogramming.

Breaking down linguistic barriers: The radical impact of translanguaging on pre-service EFL teachers' perspectives in Turkey.

PLoS One

December 2024

Department of English Language Education, Education Faculty, Mersin University, Yenişehir, Turkey.

This study examines the perceptions and practices of translanguaging among pre-service English as a Foreign Language (EFL) teachers in Turkey, employing a mixed-method research design with a phenomenological framework. The sample consists of 401 pre-service EFL teachers from the Education Faculty's English Language Teaching (ELT) Department at a state university during the 2024-2025 academic year. Data collection included a rigorously developed 28-item Likert-scale questionnaire and semi-structured interviews.

To further explore host-associated traits unique to -derived , we compared 445 high-quality genomes of available in public databases (e.

We are naturally chimeras. Apart from our own cells originating from the fertilized egg, placental mammals receive small numbers of maternal cells called maternal microchimerism (MMc) that persist throughout one's whole life. Not only are varying frequencies of MMc cells reported in seemingly contradicting phenomena, including immune tolerance and possible contribution to autoimmune-like disease, but frequencies are observable even among healthy littermates showing varying MMc frequencies and cell type repertoire.

LC-ESI-MS/MS is a preferred method for detecting and identifying metabolites, including those that are unpredictable from the genome, especially in basal metazoans like Cnidaria, which diverged earlier than bilaterians and whose metabolism is poorly understood. However, the unexpected appearance of a "ghost peak" for dopamine, which exhibited the same m/z value and MS/MS product ion spectrum during an analysis of Nematostella vectensis, a model cnidarian, complicated its accurate identification. Understanding the mechanism by which "ghost peaks" appear is crucial to accurately identify the monoamine repertoire in early animals so as to avoid misassignments.
