Chronic lymphocytic leukemia (CLL) is the most common human leukemia, and dysregulation of tRNA-derived short noncoding RNA (tsRNA) (tRF-1) expression is an accompanying event in the development of this disease. tsRNAs are fragments originating from the 3' end of tRNA precursors and do not contain mature tRNA sequences. In contrast to tsRNAs, mature tRFs (tRF-3s, tRF-5s, and internal tRFs) are produced from mature tRNA sequences and are redundant fragments. We investigated tsRNA expression in CLL and determined tsRNA signatures in indolent CLL and aggressive CLL vs. normal B cells. We noticed that both and are derived from distinct genes of pre-tRNA, and are down-regulated in CLL 3- to 5-fold vs. normal B cells. Thus, we investigated expression levels of tRF-5 fragments from tRNA in CLL samples and healthy controls, and determined that such fragments are down-regulated by 5-fold in CLLs vs. normal controls. Given these results, we investigated the expression of all mature tRFs in CLLs vs. normal controls. We found a drastic dysregulation of the expression of mature tRFs in CLL. In aggressive CLL, for the top 15 up-regulated fragments, linear fold change varied from 2,053- to 622-fold. For the top 15 down-regulated fragments in CLL, linear fold change varied from 314- to 52-fold. In addition, 964 mature tRFs were up-regulated at least 2-fold in CLL, while 701 fragments were down-regulated at least 2-fold. Similar results were obtained for indolent CLL. Our results suggest that mature tRFs may have oncogenic and/or tumor suppressor function in CLL.

Transfer RNA-derived small RNAs (tsRNAs), a recently identified non-coding RNA subset, are mainly classified into tRNA-derived small RNA fragments (tRFs) and tRNA-derived stress-induced RNAs (tiRNAs). tsRNAs dysregulation is frequently observed in numerous cancer types, suggesting involvement in tumorigenesis. However, their functions in breast cancer (BC) remain to be fully understood.

Article Synopsis
  • Sperm small RNAs, particularly tRNA fragments (tRFs), are influenced by environmental factors and play a role in passing down paternal traits.
  • The study examines the impact of four specific ribonuclease genes (Rnase9-12) on sperm fertility and small RNA levels by creating knockout (KO) mice that lack these genes.
  • KO male mice were found to be sterile, with notably low levels of tRFs in their sperm, revealing the importance of these genes in sperm development and function.

Article Synopsis
  • * Researchers investigated the role of four paralogs of Angiogenin, which are expressed in the epididymis, by creating mice that lacked these genes; the results showed that these genes are critical for male fertility and RNA processing.
  • * The KO mice were completely sterile, with their sperm unable to successfully fertilize oocytes due to impaired passage through the reproductive tract and a notable decrease in levels of tRFs, indicating these Angiogenin paralogs are vital for regulating sperm RNA composition.

tsRNA (tRNA-derived small RNA) is derived from mature tRNA or precursor tRNA (pre-tRNAs). It is lately found that tsRNA's aberrant expression is associated with tumor occurrence and development, it may be used a molecule of diagnosis and therapy. Based on the cleavage position of pre-tRNAs or mature tRNAs, tsRNAs are classified into two categories: tRNA-derived fragments (tRFs) and tRNA halves (also named tiRNAs or tRHs).

Article Synopsis
  • tRF-Val-CAC-010 is a small piece of RNA that comes from regular tRNAs and might affect how lung adenocarcinoma (a type of lung cancer) cells grow and spread.
  • Researchers found that when they blocked this tRF, the cancer cells grew less, moved less, and died more often in experiments.
  • In tests with mice, tumors were smaller in those with the tRF blocked, suggesting it helps cancer grow and spread in the body.
