Background: 3C/Ritscher-Schinzel syndrome is characterised by congenital cranio-cerebello-cardiac dysplasia, where and are accepted as the causative genes. In combination with the retromer or retriever complex, these genes play a role in endosomal membrane protein recycling. We aimed to identify the gene abnormality responsible for the pathogenicity in siblings with a 3C/Ritscher-Schinzel-like syndrome, displaying cranio-cerebello-cardiac dysplasia, coloboma, microphthalmia, chondrodysplasia punctata and complicated skeletal malformation.

Methods: Exome sequencing was performed to identify pathogenic variants. Cellular biological analyses and generation of knockout mice were carried out to elucidate the gene function and pathophysiological significance of the identified variants.

Results: We identified compound heterozygous pathogenic variants (c.1097dup; p.Cys366Trpfs*28 and c.2755G>A; p.Ala919Thr) in the gene, which encodes a core protein of the retriever complex. The identified missense variant lacked the ability to form the retriever complex, and the frameshift variant induced non-sense-mediated mRNA decay, thereby confirming biallelic loss of function of VPS35L. In addition, knockout cells showed decreased autophagic function in nutrient-rich and starvation conditions, as well as following treatment with Torin 1. We also generated mice and demonstrated that they were embryonic lethal at an early stage, between E7.5 and E10.5.

Conclusions: Our results suggest that biallelic loss-of-function variants in underlies 3C/Ritscher-Schinzel-like syndrome. Furthermore, VPS35L is necessary for autophagic function and essential for early embryonic development. The data presented here provide a new insight into the critical role of the retriever complex in fetal development.

Selective cargo and membrane recognition by SNX17 regulates its interaction with Retriever.

EMBO Rep

The Retriever complex recycles a wide range of transmembrane proteins from endosomes to the plasma membrane. The cargo adapter protein SNX17 has been implicated in recruiting the Retriever complex to endosomal membranes, yet the details of this interaction have remained elusive. Through biophysical and structural model-guided mutagenesis studies with recombinant proteins and liposomes, we have gained a deeper understanding of this process.

During endosomal recycling, Sorting Nexin 17 (SNX17) facilitates the transport of numerous membrane cargo proteins by tethering them to the Retriever complex. Despite its importance, the mechanisms underlying this interaction have remained elusive. Here, we provide biochemical, structural, cellular, and proteomic analyses of the SNX17-Retriever interaction.

Investigation of stent retriever removal forces in an experimental model of acute ischemic stroke.

Department of Mechanical, Aerospace, and Biomedical Engineering, University of Tennessee, Knoxville, TN, United States.

Introduction: Mechanical thrombectomy becomes more complex when the occlusion occurs in a tortuous cerebral anatomy, increasing the puncture to reperfusion time and the number of attempts for clot removal. Therefore, an understanding of stent retriever performance in these locations is necessary to increase the efficiency and safety of the procedure. An investigation into the effects of occlusion site tortuosity, blood clot hematocrit, and device geometry was conducted to identify their individual influence on stent retriever removal forces.

Large-scale Language Models (LLMs) utilizing the Chain-of-Thought prompting demonstrate exceptional performance in a variety of tasks. However, the persistence of factual hallucinations remains a significant challenge in practical applications. Prevailing retrieval-augmented methods treat the retriever and generator as separate components, which inadvertently restricts the generator's capabilities to those of the retriever through intensive supervised training.

Background: Distal embolization is a frequent complication of mechanical thrombectomy (MT) for acute ischemic stroke, often leading to poor clinical outcomes. The vascular bifurcations represent a specialized anatomical location, thereby augmenting the complexity of MT. The specific factors contributing to distal embolization in this context have not been thoroughly explored.
