A series of novel quinolinone derivatives bearing dithiocarbamate moiety were designed and synthesised as multifunctional AChE inhibitors for the treatment of AD. Most of these compounds exhibited strong and clearly selective inhibition to AChE. Among them, compound was identified as the most potent inhibitor to both AChE and AChE (IC = 0.22 μM for eeAChE; IC = 0.16 μM for AChE), and it was also the best inhibitor to AChE-induced Aβ aggregation (29.02% at 100 μM) and an efficient inhibitor to self-induced Aβ aggregation (30.67% at 25 μM). Kinetic and molecular modelling studies indicated that compound was a mixed-type inhibitor, which could interact simultaneously with the catalytic anionic site (CAS) and the peripheral anionic site (PAS) of AChE. In addition, had good ability to cross the BBB, showed no toxicity on SH-SY5Y neuroblastoma cells and was well tolerated in mice at doses up to 2500 mg/kg (po).

Aryl quinolone derivatives can target the cytochrome bc complex of Plasmodium falciparum, exhibiting excellent in vitro and in vivo antimalarial activity. However, their clinical development has been hindered due to their poor aqueous solubility profiles. In this study, a series of bioisosteres containing saturated heterocycles fused to a 4-pyridone ring were designed to replace the inherently poorly soluble quinolone core in antimalarial quinolones with the aim to reduce π-π stacking interactions in the crystal packing solid state, and a synthetic route was developed to prepare these alternative core derivatives.

Background: For the treatment of gastritis, rebamipide, a mucoprotective agent, and nizatidine, a gastric acid suppressant, are commonly employed individually.
Aim: To compare the efficacy of Mucotra SR (rebamipide 150 mg) and Axid (nizatidine 150 mg) combination therapy with the sole administration of Axid in managing erosive gastritis.
Methods: A total of 260 patients diagnosed with endoscopically confirmed erosive gastritis were enrolled in this open-label, multicenter, randomized, phase 4 clinical trial, allocating them into two groups: Rebamipide/nizatidine combination twice daily nizatidine twice daily for 2 weeks.

Sorption and transport are important environmental behaviors of antibiotics in soils and can determine the fate of antibiotics in environments; however, limited relevant studies have been conducted on long-term manured soils. In this study, batch and repacked soil column experiments were conducted to examine the sorption and transport behavior of four veterinary antibiotics, including sulfamethazine (SMT), florfenicol (FFC), doxycycline (DOX), and enrofloxacin (ENR), in red soils, yellow soils, and calcareous soils with long-term amendment of chicken or pig manure collected in Zhejiang Province. The results showed that the sorption isothermal data of the four target antibiotics all conformed well to the linear and Freundlich models.
