Laccase (EC 1.10.3.2; benzenediol; oxygen oxidoreductases) is a multi-copper oxidase that catalyzes the oxidation of phenols, polyphenols, aromatic amines, and different non-phenolic substrates with concomitant reduction of O to HO. Enzymatic oxidation techniques have the potential of implementation in different areas of industrial fields. In this study, the sp. A01 laccase gene was cloned into pET-26 (b+) vector and was transformed to BL21. Then it was purified using His tag affinity (Ni sepharose resin) chromatography. The estimated molecular weight was approximately 60 kDa using SDS-PAGE. The highest enzyme activity and best pH for 2,6-dimethoxyphenol (DMP) oxidation were recorded as 8 at 90 °C respectively. The calculated half-life and kinetic values including K, V, turn over number (k), and catalytic efficiency (k/K) of the enzyme were 106 min at 90 °C and 686 μM, 10.69 U/ml, 20.3 S, and 0.029 s μM, respectively. The DMP was available as the substrate in all the calculations. Enzyme activity enhanced in the presence of Cu, NaCl, SDS, n-hexane, Triton X-100, tween 20, and tween 80, significantly. The binding residues were predicted and mapped upon the modeled tertiary structure of identified laccase. The remaining activity and structural properties of sp. A01 laccase in extreme conditions such as high temperatures and presence of metals, detergents, and organic solvents suggest the potential of this enzyme in biotechnological and industrial applications. This process has been patented in Iranian Intellectual Property Centre under License No: 91325.

Seed size is not only a yield-related trait but also an important measure to determine the commercial value of groundnut in the international market. For instance, small size is preferred in oil production, whereas large-sized seeds are preferred in confectioneries. In order to identify the genomic regions associated with 100-seed weight (HSW) and shelling percentage (SHP), the recombinant inbred line (RIL) population (Chico × ICGV 02251) of 352 individuals was phenotyped for three seasons and genotyped with an Axiom_ array containing 58K SNPs.

Sustainable use of the spent mushroom substrate of Pleurotus florida for production of lignocellulolytic enzymes.

J Basic Microbiol

February 2020

ICAR-National Bureau of Agriculturally Important Microorganisms, Mau, Uttar Pradesh, India.

Spent mushroom substrate (SMS), a major byproduct of the mushroom industry, is a lignocellulosic biomass, which contains approximately 57-74.3% of holocellulose fraction. This study was aimed at utilizing SMS of Pleurotus florida for recovery of lignocellulolytic enzymes and sugars and also as a substrate for production of cellulolytic enzymes using different isolates of Trichoderma and Aspergillus under solid-state fermentation (SSF).

Laccase-TEMPO-mediated air oxidation of galactomannan for use as paper strengthening agent.

Carbohydr Polym

March 2018

MOE Engineering Research Center of Forestry Biomass Materials and Bioenergy, Beijing Forestry University, Beijing, 100083, China. Electronic address:

In this study, galactomannans, which are a type of plant polysaccharide, were oxidized by using a laccase and TEMPO-mediated air-oxidation system. The influence of the oxidation conditions, including the laccase doses, oxidation times, and amounts of oxidizing system, on the properties of the paper were investigated. The results showed that the oxidized galactomannans could improve the paper properties.

Article Synopsis
  • A laccase-based biosensor was developed to determine polyphenol content in in vitro Salvia cultures, focusing on measuring rosmarinic acid equivalent content.
  • The biosensor utilized a specific construction method to ensure accurate electrochemical measurements, achieving a low limit of detection and a defined linear response range.
  • It proved to be stable and sensitive, effectively monitoring total polyphenolic content without interference from other substances in the growth medium.
