Objective: Clinical trials have demonstrated the efficacy of indigo naturalis, a traditional Chinese medicine ingredient, against psoriasis, a skin disease characterized by keratinocyte hyperproliferation and inflammation. The present study investigates the efficacy of tryptanthrin, a bioactive compound in indigo naturalis, against non-melanoma skin cancer (NMSC) and the signalling events involved.

Methods: Efficacy of tryptanthrin against NMSC was assessed using DMBA/PMA-induced skin carcinogenesis model in Swiss albino mice. Immunostaining for PCNA and ki-67 was used to mark proliferating cells in tissues. Haematoxylin and eosin staining and toluidine staining were employed to assess inflammation, and TUNEL assay was used to detect apoptosis in tissues. The signalling events were evaluated using Western blot, imunohistochemistry and immunofluorescence staining. MTT assay and clonogenic assay were performed to assess the viability and proliferation of cancer cells, in vitro.

Results: In mice, topical application of tryptanthrin suppressed skin carcinogenesis. It attenuated inflammation, impeded the proliferation of hair follicle (HF) cells and suppressed the activation of β-catenin, a major driver of HF cell proliferation. Additionally tryptanthrin suppressed the activation of ERK1/2 and p38, both of which promote β-catenin activation and lowered the expression of c-Myc and cyclin-D1. Tryptanthrin suppressed the proliferation of the human NMSC cell line, A431 and abrogated EGF-induced activation of β-catenin and subsequent cytoskeletal rearrangement.

Conclusion: The study demonstrates with molecular evidence that tryptanthrin is an effective suppressor of NMSC.

Article Synopsis
  • Ulcerative colitis (UC) is a challenging inflammatory bowel disease with limited treatment options, prompting the need for safer and more effective drugs.
  • Recent studies show that 'Indigo naturalis' (IN), a traditional Chinese medicine, has strong anti-inflammatory, antioxidant, and immunomodulatory effects, making it a promising candidate for UC treatment.
  • Key components like indirubin and indigo from IN have been shown to effectively treat severe UC, but potential risks such as pulmonary hypertension highlight the need for careful use and further research on its therapeutic value.

Background/objectives: Isatidis Folium (IF) has been used in traditional medicine for various ailments, and recent research highlights its anti-inflammatory, antiviral, and detoxifying properties. This study investigated the anti-inflammatory effects of a hydroethanolic extract of IF (EIF) on inflammasomes and colitis.

Methods: Dextran sulfate sodium (DSS)-induced colitis model C57BL/6 mice were treated with DSS, mesalamine, or EIF (200 mg/kg).

Article Synopsis
  • - This study explores the potential of IN compounds, particularly indirubin and indigo, as a novel treatment for Inflammatory Bowel Disease (IBD) by analyzing their pharmacological effects and mechanisms in preclinical models.
  • - It evaluates important parameters such as Disease Activity Index (DAI) score, colon length, and mucosal damage in mice with DSS-induced colitis, while also investigating anti-inflammatory pathways like AhR, NF-κB, and TLR4.
  • - Findings indicate that IN significantly improves symptoms of IBD and positively impacts intestinal microbiota diversity, suggesting it could be an effective therapeutic option for managing the disease.

Background: The adaptor protein apoptosis-associated speck-like protein (ASC) containing a caspase recruitment domain (CARD) can be activated through pyrin domain (PYD) interactions between sensors and ASC, and through CARD interactions between caspase-1 and ASC. Although the majority of ternary inflammasome complexes depend on ASC, drugs targeting ASC protein remain scarce. After screening natural compounds from Isatidis Radixin, we found that tryptanthrin (TPR) could inhibit NLRP3-induced IL-1β and caspase-1 production, but the underlying anti-inflammatory mechanisms remain to be elucidated.
