A palladium-catalyzed uniquely regioselective C-H alkynylation of indoles has been described. In this protocol, simple and readily available haloalkynes are employed as efficient alkynylating reagents, affording a series of functionalized 7-alkynylindoles in moderate to good yields. Moreover, further transformations of 7-alkynylated products were performed, which demonstrated the potential application of this method in organic synthesis.

ConspectusIn recent years, our research group has dedicated significant effort to the field of asymmetric organometallic electrochemical synthesis (AOES), which integrates electrochemistry with asymmetric transition metal catalysis. On one hand, we have rationalized that organometallic compounds can serve as molecular electrocatalysts (mediators) to reduce overpotentials and enhance both the reactivity and selectivity of reactions. On the other hand, the conditions for asymmetric transition metal catalysis can be substantially improved through electrochemistry, enabling precise modulation of the transition metal's oxidation state by controlling electrochemical potentials and regulating the electron transfer rate via current adjustments.

Synthesis of 2-Sulfonyl Carbazoles via Oxidative C-H Functionalization of Tetrahydrocarbazoles with Sulfonyl Hydrazides.

Herein, we report an approach for the synthesis of 2-sulfonyl carbazoles from the oxidative C-H sulfonylation of tetrahydrocarbazoles. The mechanistic study reveals that this special selectivity is realized by the addition of a sulfonyl radical to the 3,4-dihydrocabazole intermediate via dehydrogenative desaturation of tetrahydrocarbazoles. This approach features readily available starting materials, high regioselectivity, broad substrate scope, and attractive synthetic utility.

Regioselective Difluoroalkylation of 2-Pyridones with Fluoroalkyl Bromides Enabled by a Nickel(II) Catalyst.

Regioselective C-H difluoroalkylation of diverse 2-pyridones with ethyl bromodifluoroacetates and bromodifluoroacetamides is accomplished by using a (dppf)NiCl catalyst under mild conditions. This efficient protocol could deliver a variety of C-3 difluoroalkylated pyridones with the tolerance of a range of highly susceptible functionalities, such as -Cl, -Br, -I, -COMe, -CN, -NMe and -NO, including heteroarenes like pyridinyl, furanyl, thiophenyl and carbazolyl moieties. A preliminary mechanistic study suggests the radical pathway for the reaction involving fluoroalkyl radical intermediate.
