The relationship between the N-methylformamide (NMF)-induced differentiation of HL-60 human promyelocytic leukemia cells, to myeloid cells, and their proliferative potential was determined in suspension culture. Continuous incubation with 170 mM NMF induced 70% of cells to differentiate after 2-2.5 cell divisions, whether or not cells were replenished with fresh medium and serum. 100 mM NMF induced significant levels of differentiation only when cells were allowed to reach a plateau of growth, and this was prevented by replenishment of the medium and serum. Under these conditions of "re-feeding", the cells could be made to undergo greater than 10 cell divisions, and differentiated only when they were allowed to reach a plateau in their growth. The data suggests that NMF may induce differentiation only when the proliferative potential of the cells is limited, either by the drug itself or by the conditions of growth.

In many industrial activities, workers may be exposed by inhalation to particles that are aerosolized, To predict the human health hazard of these materials, we propose to develop a co-culture model (macrophages, granulocytes, and alveolar epithelial cells) designed to be more representative of the inflammatory pulmonary response occurring in vivo. Phorbol 12-myristate 13-acetate (PMA)-differentiated THP-1 cells were used as macrophages, All-trans retinoic acid (ATRA)-differentiated HL60 were used as granulocytes and A549 were used as epithelial alveolar type II cells. A crystalline silica sample DQ12 was used as a prototypical particle for its capabilities to induce DNA damage, inflammatory response, and oxidative stress in epithelial cells; its polyvinylpyridine-N-oxide (PVNO)-surface modified counterpart was also used as a negative particulate control.

Oncostatin M (OSM) plays a crucial role in diverse inflammatory reactions. Although the food bioactive compound naringenin (NAR) exerts various useful effects, including antitussive, anti-inflammatory, hepatoprotective, renoprotective, antiarthritic, antitumor, antioxidant, neuroprotective, antidepressant, antinociceptive, antiatherosclerotic, and antidiabetic effects, the modulatory mechanism of NAR on OSM expression in neutrophils has not been specifically reported. Leukopenia, marked by diminished white blood cell (WBC) counts, presents significant challenges in the management of hematological malignancies and immunocompromised patients. This study evaluated the therapeutic potential of miltefosine (MFS), a phospholipid analogue, for treating leukopenia. In vitro studies using HL60 and NB4 cells revealed that MFS effectively promoted neutrophil differentiation and function, evidenced by the upregulation of surface markers CD11b, CD11c, CD14, and CD15, as well as enhanced bactericidal activity assessed through the NBT reduction assay.
