Theories about the origin of life require chemical pathways that allow formation of life's key building blocks under prebiotically plausible conditions. Complex molecules like RNA must have originated from small molecules whose reactivity was guided by physico-chemical processes. RNA is constructed from purine and pyrimidine nucleosides, both of which are required for accurate information transfer, and thus Darwinian evolution. Separate pathways to purines and pyrimidines have been reported, but their concurrent syntheses remain a challenge. We report the synthesis of the pyrimidine nucleosides from small molecules and ribose, driven solely by wet-dry cycles. In the presence of phosphate-containing minerals, 5'-mono- and diphosphates also form selectively in one-pot reactions. The pathway is compatible with purine synthesis, allowing the concurrent formation of all Watson-Crick bases.

Long COVID and gut microbiome: insights into pathogenesis and therapeutics.

Gut Microbes

Post-acute coronavirus disease 2019 syndrome (PACS), following severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection or coronavirus disease 2019 (COVID-19), is typically characterized by long-term debilitating symptoms affecting multiple organs and systems. Unfortunately, there is currently a lack of effective treatment strategies. Altered gut microbiome has been proposed as one of the plausible mechanisms involved in the pathogenesis of PACS; extensive studies have emerged to bridge the gap between the persistent symptoms and the dysbiosis of gut microbiome.

Concomitant formation of protocells and prebiotic compounds under a plausible early Earth atmosphere.

Revealing the origin of life and unambiguously detecting fossil remains of the earliest organisms are closely related aspects of the same scientific research. The synthesis of prebiotic molecular building blocks of life and the first compartmentalization into protocells have been considered two events apart in time, space, or both. We conducted lightning experiments in borosilicate reactors filled with a mixture of gases mimicking plausible geochemical conditions of early Earth.

ConspectusOrigin of Life research is a fast growing field of study with each year bringing new breakthroughs. Recent discoveries include novel syntheses of life's building blocks, mechanisms of activation and interaction between molecules, and newly identified environments that provide promising conditions for these syntheses and mechanisms. Even with these new findings, firmly grounded in rigorous laboratory experiments, researchers often find themselves uncertain about how to apply them.

Protein-free catalysis of DNA hydrolysis and self-integration by a ribozyme.

Group II introns are ancient self-splicing ribozymes and retrotransposons. Though long speculated to have originated before translation, their dependence on intron-encoded proteins for splicing and mobility has cast doubt on this hypothesis. While some group II introns are known to retain part of their catalytic repertoire in the absence of protein cofactors, protein-free complete reverse splicing of a group II intron into a DNA target has never been demonstrated.

Prebiotic chemistry and protocells during the faint young sun event.

Biophys Rev

Space Science Center (ANGKASA), Institute of Climate Change, National University of Malaysia, 43600 Selangor, Malaysia.

The sun generates light and heat for life on Earth to flourish. However, during the late Hadean-early Archean epoch on Earth, the "faint young sun" (FYS) was less luminous, influencing prebiotic chemistry and, by extension, the origins of life (OoL). However, higher levels of ultraviolet (UV) radiation from the FYS, especially UV-C, due to the lack of an ozone layer, would likely have impacted the assembly, stability, persistence, and functions of prebiotic cellular precursors, i.
