South American Zika virus (ZIKV) recently emerged as a novel human pathogen, linked with neurological disorders. However, comparative ZIKV infectivity studies in New World primates are lacking. Two members of the Callitrichidae family, common marmosets (Callithrix jacchus) and red-bellied tamarins (Saguinus labiatus), were highly susceptible to sub-cutaneous challenge with the Puerto Rico-origin ZIKV strain. Both exhibited rapid, high, acute viraemia with early neuroinvasion (3 days) in peripheral and central nervous tissue. ZIKV RNA levels in blood and tissues were significantly higher in New World hosts compared to Old World species (Macaca mulatta, Macaca fascicularis). Tamarins and rhesus macaques exhibited loss of zonal occludens-1 (ZO-1) staining, indicative of a compromised blood-brain barrier 3 days post-ZIKV exposure. Early, widespread dissemination across multiple anatomical sites distant to the inoculation site preceded extensive ZIKV persistence after 100 days in New and Old World lineages, especially lymphoid, neurological and reproductive sites. Prolonged persistence in brain tissue has implications for otherwise resolved human ZIKV infection. High susceptibility of distinct New World species underscores possible establishment of ZIKV sylvatic cycles in primates indigenous to ZIKV endemic regions. Tamarins and marmosets represent viable New World models for ZIKV pathogenesis and therapeutic intervention studies, including vaccines, with contemporary strains.

Flaviviruses utilize the cellular endoplasmic reticulum (ER) for all aspects of their lifecycle. Genome replication and other viral activities take place in structures called replication organelles (ROs), which are invaginations induced in the ER membrane. Among the required elements for RO formation is the biogenesis of viral nonstructural proteins NS4A and NS4B.

Article Synopsis
  • In-utero exposure to Zika virus (ZIKV) can result in severe outcomes like miscarriage and congenital Zika syndrome, prompting a study on the infection rates among pregnant women from February to April 2022.
  • Researchers tested 200 pregnant women for ZIKV antibodies and found that 16.5% had ZIKV IgM, 10% had IgG, and 23% had neutralizing antibodies, indicating various stages of infection and immunity.
  • The study also revealed that recent ZIKV infections were more common in sexually active women aged 20-29, with higher infection risks in first trimester and grand-multiparous women, although no significant link was found between ZIKV and specific pregnancy trimest

Rapidly identifying Anopheles-carrying malaria parasites is crucial for imported malaria prevention. However, suitable methods still lack quick detection in limited-resource situations. In this study, disc microfluidic isothermal amplification integrating loop-mediated isothermal amplification (LAMP) and microfluidic chip technology were applied to develop rapid and precise detection with low resource requirements.

Public Health and community medicine Department, Theodor Bilharz Research Institute, Helwan University, Cairo, Egypt.

Infectious diseases significantly impact both public health and economic stability, underscoring the critical need for precise outbreak predictions to effictively mitigate their impact. This study applies advanced machine learning techniques to forecast outbreaks of Dengue, Chikungunya, and Zika, utilizing a comprehensive dataset comprising climate and socioeconomic data. Spanning the years 2007 to 2017, the dataset includes 1716 instances characterized by 27 distinct features.

Epidemic Zika virus strains from the Asian lineage induce an attenuated fetal brain pathogenicity.

Nat Commun

December 2024

KU Leuven Department of Microbiology, Immunology and Transplantation, Virology, Antiviral Drug & Vaccine Research Group, Rega Institute for Medical Research, Leuven, Belgium.

The 2015-2016 Zika virus (ZIKV) outbreak in the Americas revealed the ability of ZIKV from the Asian lineage to cause birth defects, generically called congenital Zika syndrome (CZS). Notwithstanding the long circulation history of Asian ZIKV, no ZIKV-associated CZS cases were reported prior to the outbreaks in French Polynesia (2013) and Brazil (2015). Whether the sudden emergence of CZS resulted from an evolutionary event of Asian ZIKV has remained unclear.
