In the daily diet, sweeteners play an indispensable role. Among them, sucrose, a widely occurring disaccharide in nature, is a commonly used sweetener. However, the intake of sucrose can cause a rapid increase in blood glucose, which leads to a number of health problems. Therefore, there is an urgent need for possible alternatives to sucrose. Currently, four naturally occurring sucrose isomers, trehalulose, turanose, leucrose, and isomaltulose are considered to be possible alternatives to sucrose due to their suitable sweetness, potential physiological benefits, and feasible production processes. This review covers the properties of these alternative sweeteners, including their structure, sweetness, hydrolysis rate, toxicology, and cariogenicity, and exhibits their potential applications in chronic diseases management, anti-inflammatory supplement, prebiotic dietary supplement, and stabilizing agent. The biosynthesis of these sucrose isomers using carbohydrate-active enzymes and their industrial production processes are also systematically summarized.

Rebaudioside M2 (RebM2) is characterized as 13-[(2--β-d-glucopyranosyl-3--β-d-glucopyranosyl-β-d-glucopyranosyl)oxy] ent-kaur-16-en-19-oic acid-[(2--β-d-glucopyranosyl-6--β-d-glucopyranosyl-β-d-glucopyranosyl) ester], an isomer of rebaudioside M with a 1 → 6 sugar linkage. The product was found in the biotransformation of rebaudioside D (RebD) catalyzed by a glycosyltransferase from (UGT). Herein, guided by consensus engineering and molecular dynamics simulations, a variant UGT with enhanced activity and thermostability was obtained.

Turanose (α-d-glucopyranosyl-(1→3)-α-d-fructose) is a rare disaccharide that is a potential low-calorigenic sweetener. Anhedonia and avolition are emotions frequently endorsed by individuals with stress related disorders. Kappa opioid receptor (KOR) activation can induce negative emotions and recent clinical evidence suggests that KOR antagonism can alleviate anhedonia in a transdiagnostic cohort of patients. However, the behavioral consequences of KOR activation and antagonism in modulating motivation, as assessed by schedule-controlled behavioral performance without preexisting conditions (stress or substance use), have not been formally assessed.
