Inhibition of Amyloid-β Aggregation by Cobalt(III) Schiff Base Complexes: A Computational and Experimental Approach.

Coordination complexes have emerged as prominent modulators of amyloid aggregation via their interaction with the N-terminal histidine residues of amyloid-β (Aβ). Herein, we report the synthesis and characterization of a novel cobalt(III) Schiff base complex with methylamine axial ligands, and we present both computational and experimental data demonstrating the reduction of β-sheet formation by this complex. The computations include molecular dynamics simulations of both monomeric and pentameric Aβ, which demonstrate decreased formation of β-sheet structures, destabilization of preformed β-sheets, and suppression of aggregation. These results are consistent with a dose dependence in experimental bulk aggregation data using thioflavin T fluorescence, and overall this study demonstrates useful drug activity of the cobalt complex.

A new one-dimensional thiocyanate-bridged cobalt(III) Schiff base complex, namely, catena-poly[[[4-bromo-2-((Z)-{[2-(thiophen-2-yl)ethyl]imino}methyl)phenolato-κN,O]cobalt(III)]-μ-thiocyanato-κN:S], [Co(SCN)(CHBrNOS)] or [Co(μ-SCN)L] (1), where HL is 4-bromo-2-((Z)-{[2-(thiophene-2-yl)ethyl]imino}methyl)phenol, a bidentate Schiff base prepared from the condensation reaction of 5-bromosalicylaldehyde and 2-(thiophen-2-yl)ethylamine, has been synthesized by stirring Co(ClO)·6HO, the Schiff base HL and ammonium thiocyanate (in a 1:2:1 molar ratio) in ethanol medium. The complex was characterized by FT-IR, electronic spectra and single-crystal X-ray diffraction (SC-XRD) studies. The SC-XRD data suggest that the compound crystallizes in the orthorhombic space group Pca2.

A set of mononuclear cobalt(III) octahedral complexes {[Co(L)(acac)] (Co-1H), [Co(L)(acac)] (Co-1Br), and [Co(L)(acac)] (Co-1NO2)} were synthesized using new-generation N/O donors, maleonitrile-tethered, tetradentate heteroscorpionate half-reduced Schiff base ligands, 2-(()-2-hydroxybenzylideneamino)-3-(pyridin-2-ylmethylamino)maleonitrile (H2LH), 2-(()-(5-bromo-2-hydroxybenzylidene)amino)-3-((pyridin-2-ylmethyl)amino)maleonitrile (H2LBr), and 2-(()-2-hydroxy-5-nitrobenzylideneamino)-3-(pyridin-2-ylmethylamino)maleonitrile (H2LNO2). All the compounds were well characterized spectroscopically and structurally. The non-covalent interactions present in the lattice of Co-complexes were studied in detail to explain the molecular architecture using the Hirshfeld surface (HS) analysis.

Three cobalt complexes, namely [Co(HL)(N)]ClO (1), [Co(L)(HL)(N)]ClO·1.5HO (2), and [Co(L)(HL)(NCS)] [CoCl(NCS)] (3), where HL = 2-(3-(dimethylamino)propyliminomethyl)-6-methoxyphenol, HL = 2-(2-(dimethylamino)ethyliminomethyl)-4,6-dichlorophenol, and HL = 2-(2-(dimethylamino)ethyliminomethyl)-6-methoxyphenol, as potential tridentate NO-donor Schiff base ligands, were synthesized and characterized using elemental analysis, IR and UV-vis spectroscopy, and single-crystal X-ray diffraction studies. All three were found to be monomeric ionic complexes.

Two Schiff base complexes of copper(ii) and cobalt(iii) having the formulae [CuL] (Cu-Sal) and [CoL] (Co-Sal) (HL = 2-(((2-hydroxyethyl)imino)methyl)phenol) have been synthesized and characterized microanalytically, spectroscopically and in the case of Cu-Sal using single crystal X-ray diffraction technique. The single crystal X-ray analysis reveals a square planar geometry around Cu(ii) satisfied by phenoxide oxygen and imine nitrogen of the L ligand to generate a six membered chelate ring. The solid state structure of Cu-Sal is satisfied by varied intermolecular non-covalent interactions.

A bis-Schiff base N,N'-ethylene-bis(3-bromosalicylaldimine) (H2L) was prepared from 3-bromosalicylaldehyde and ethane-1,2-diamine. With H2L as ligand, a new copper(II) complex [CuL] (1) and a new cobalt(III) complex [CoL(NCS)(DMF)] (2) were prepared and characterized by physico-chemical methods and single crystal X-ray analysis. X-ray analysis indicates that the Cu atom in complex 1 is in square planar coordination, and the Co atom in complex 2 is in octahedral coordination.
