Salicylic acid (SA) is an important signaling hormone in plant immunity. It can be synthesized by either the phenylpropanoid pathway or the isochorismate pathway, but mutant studies of this have been scarce in other species than Arabidopsis. Here we identified a mutation that introduced a stop-codon early in the barley gene for isochorismate synthase (ICS). We found that homozygous plants wilted if not sprayed with 1,4-dihydroxy-2-naphthoic acid, a precursor of phylloquinone, also synthesized via the isochorismate pathway. Interestingly, had unchanged SA, suggesting that the basal level of SA is synthesized via the phenylpropanoid pathway. Previous studies have failed seeing increased SA levels in barley after attack by the powdery mildew fungus, f.sp. (), and indeed, we saw no changes in the interaction of with this fungus. Overall, we hope this mutant will be useful for other studies of SA in barley.

Plants subjected to cold stress have been observed to accumulate proline, but the underlying regulatory mechanism remains to be elucidated. In this study, we identified a pyrroline-5-carboxylate synthetase (P5CS)-encoding gene (CtrP5CS1) from trifoliate orange (Citrus trifoliata L.), a cold-hardy citrus species, as a critical gene for cold-induced proline accumulation.

Article Synopsis
  • Salicylic acid (SA) is a crucial plant hormone involved in growth and defense, produced mainly through two pathways: isochorismate synthase (ICS) and phenylalanine ammonia-lyase (PAL).
  • Researchers used the CRISPR/Cas9 technology to create knockout mutants in rice that showed significant growth issues and lacked phylloquinone, suggesting a direct connection between OsICS and phylloquinone synthesis.
  • Despite the growth defects, the levels of SA in the mutants remained unchanged, indicating that rice may utilize a different pathway for SA biosynthesis.

Vascular plant one-zinc finger (VOZ) transcription factors (TFs) play crucial roles in plant immunity. Nevertheless, how VOZs modulate defense signaling in response to elicitor-induced resistance is not fully understood. Here, the defense elicitor β-aminobutyric acid (BABA) resulted in the visible suppression of Rhizopus rot disease of peach fruit caused by Rhizopus stolonifer.

Article Synopsis
  • * This research identifies the rapid transcriptional changes that occur shortly after NHP treatment, revealing that WRKY transcription factor genes respond first, followed by defense-related genes in a secondary response.
  • * The study also finds that while the initial response relies on low salicylic acid (SA) levels, the later response does not depend on new SA production, indicating the importance of WRKY70 in regulating these gene expressions related to SAR and reactive oxygen species (ROS) production.

In plants, a local infection can lead to systemic acquired resistance (SAR) through increased production of salicylic acid (SA). For many years, the identity of the mobile signal and its direct transduction mechanism for systemic SA synthesis in initiating SAR have been debated. We found that in , after a local infection, the conserved cysteine residue of the transcription factor CCA1 HIKING EXPEDITION (CHE) undergoes sulfenylation in systemic tissues, which enhances its binding to the promoter of the SA-synthesis gene () and increases SA production.
