AI Article Synopsis

  • Apocynin is a natural compound with various biological activities, leading to the design and synthesis of new derivatives for potential therapeutic use.
  • These derivatives were tested for their ability to cross the blood-brain barrier, their effects on reactive oxygen species (ROS), and their anti-glioma properties.
  • The compound D31 showed promising results by effectively penetrating the BBB, increasing ROS levels, and inhibiting glioma growth through the suppression of the NF-κB pathway, suggesting its potential for future anti-glioma treatments.

Article Abstract

Apocynin (4-hydroxy-3-methoxyacetophenone) is a natural polyphenolic compound with multiple biological activities. In the present study, a series of apocynin derivatives were designed and synthesized. The ADMET prediction, blood-brain barrier (BBB) penetration assay, anti-NADPH oxidase activity, reactive oxygen species (ROS) levels, and anti-glioma effects of these apocynin derivatives were evaluated. The anti-glioma mechanisms of candidate compounds were studied by flow cytometer and Western blot. The results showed that D31 exhibited higher BBB penetration, increased ROS generations and significant anti-glioma effects both and . Further studies showed that D31 inhibited the activations of NF-κB pathway. Overall, our data demonstrated that D31 inhibited growth and induced apoptosis of glioma, which might be caused by ROS-related NF-κB activation. The current study suggested that D31 could be further explored for its potential use in anti-glioma therapy.

Introduction: Lignin is a promising resource for obtaining aromatic materials, however, its heterogeneous structure poses a challenge for effective utilization. One approach to produce homogeneous aromatic materials from lignin involves the application of microbial catabolism, which is gaining attention. This current study focused on constructing a catabolic pathway in Pseudomonas sp.

Objective: Herein, we investigate the potential analgesic effect of a newly synthesized chalcone-derived apocynin in a neurogenic pain model.

Methods: Molecular docking was used to foretell the apocynin binding features and dynamics with the TRPV1 channel, and the activity was tested in vitro, using transfected HEK 293T cells with the rat TRPV1 receptor. The analgesic effect of apocynin was investigated using a capsaicin-induced pain model.

Human and Environmental Physiology Research Unit, School of Human Kinetics, University of Ottawa, Ottawa, Ontario, Canada.

We evaluated reactive oxygen species (ROS) modulation of cutaneous vasodilation during local and whole body passive heating in young and older adults. Cutaneous vascular conductance normalized to maximum vasodilation (%CVC) was assessed in young and older adults (10/group) using laser-Doppler flowmetry at four dorsal forearm sites treated with ) Ringer solution (control), ) 100 µM apocynin (NADPH oxidase inhibitor), ) 10 µM allopurinol (xanthine oxidase inhibitor), or ) 10 µM tempol (superoxide dismutase mimetic), via intradermal microdialysis during local () and whole body heating (). In , forearm skin sites were set at 33°C during baseline and then progressively increased to 39°C and 42°C (30 min each).

Amyotrophic lateral sclerosis (ALS) is a fatal disease that causes degeneration of motor neurons (MNs) and paralysis. ALS can be caused by mutations in the gene that encodes copper/zinc superoxide dismutase (SOD1). SOD1 is known mostly as a cytosolic antioxidant protein, but SOD1 is also in the nucleus of non-transgenic (tg) and human SOD1 (hSOD1) tg mouse MNs.
