An ultra-sensitive and "turn-on" method is demonstrated for the determination of uranyl ion. The assay is based on hairpin-to-DNAzyme structure switching that is induced by an entropy-driven catalytic reaction. An UO-specific DNAzyme is cleaved by UO to produce a DNA fragment. This fragment initiates the entropy-driven catalytic reaction to produce a large number of a sequence "R". The sequence R initiates the circular cleavage of FAM-labeled hairpins by switching the hairpin to Mg-specific DNAzyme structure. This causes the recovery of green fluorescence. The method works in the 20 pM to 800 pM concentration range and the limit of detection is 4 pM. The rapid, sensitive and reliable detection of oral cancer overexpressed 1 (ORAOV 1) is crucial for the early, non-invasive diagnosis of oral squamous cell carcinoma (OSCC). Herein, we are the first to construct an ultrasensitive electrochemical (EC) biosensor based on an entropy-driven "two-way signal output" (TWSO) cyclic circuit for salivary ORAOV 1 detection. This innovative TWSO cyclic circuit can skillfully convert by-products into desired signal-generating units, not only reducing the excessive accumulation of by-products but also improving the utilization efficiency of output chains, thereby achieving rapid reaction kinetics and high signal outputs.

The development of B-lymphoblastic leukemia is tightly associated with aberrant expression of Pax-5a. This work presented a novel dual signal amplification strategy-based Pax-5a detection method by combining the rolling circle amplification reaction (RCA) and the Entropy-driven toehold-mediated strand displacement (ETSD). Particularly noteworthy is the employed ETSD, which effectively improves the rate and stability of the reaction due to its unique entropy-driven principle.
