Plum pox virus, the agent that causes sharka disease, is among the most important plant viral pathogens, affecting trees across the globe. The fabric of interactions that the virus is able to establish with the plant regulates its life cycle, including RNA uncoating, translation, replication, virion assembly, and movement. In addition, plant-virus interactions are strongly conditioned by host specificities, which determine infection outcomes, including resistance. This review attempts to summarize the latest knowledge regarding -host interactions, giving a comprehensive overview of their relevance for viral infection and plant survival, including the latest advances in genetic engineering of resistant species.

We report an 18-year-old male who presented with a two-month history of a lesion over his right forearm with a one-week history of sudden increase in size associated with pain. General and systemic examinations were normal. Dermatological examination revealed a single tender, well-defined, pearly white to erythematous, dome-shaped nodule of approximately 6mm x 5mm x 5mm with central umbilication and surrounding erythema.

Orf is a parapoxvirus endemically found in sheep and goats. Orf can be transmitted to humans in close contact with infected animals, causing a self-limited viral skin infection known as ecythyma contagiosum. The diagnosis can be made clinically and treatment is usually conservative; however, unfamiliarity with orf may lead to an extended work-up.

Viral adhesion to host cells is a critical step in infection for many viruses, including monkeypox virus (MPXV). In MPXV, the H3 protein mediates viral adhesion through its interaction with heparan sulfate (HS), yet the structural details of this interaction have remained elusive. Using AI-based structural prediction tools and molecular dynamics (MD) simulations, we identified a novel, positively charged α-helical domain in H3 that is essential for HS binding.
