This article describes the simultaneous determination of bedaquiline fumarate (TMC-207) and rifabutin in human plasma by stable isotope dilution tandem mass spectrometry. The methodology was developed for an investigation of potential drug-drug interactions of the two anti-tuberculosis drugs when given together to healthy human volunteers. Use of the two drugs in combination to treat disease caused by Mycobacterium tuberculosis is contemplated as the bacterium becomes resistant to many currently available drugs.

Macrophages are important mediators of immune responses with critical roles in the recognition and clearance of pathogens, as well as in the resolution of inflammation and wound healing. The neuronal guidance cue SLIT2 has been widely studied for its effects on immune cell functions, most notably directional cell migration. Recently, SLIT2 has been shown to directly enhance bacterial killing by macrophages, but the effects of SLIT2 on inflammatory activation of macrophages are less known.

Antibodies in human milk protect infants against infections, but currently no assay is described that is able to simultaneously measure all 9 antibody isotypes and subclasses immunoglobulins in human fluids, such as human milk. Our cohort "Protecting against Respiratory tract Infections through human Milk Analysis" (PRIMA) is focused on the relation between the occurrence of respiratory infections during the first year of life and concentration of maternal antibodies in breastfeeding. We developed and successfully validated a multiplex assay that is able to measure all nine antibody isotypes and subclasses in human plasma and milk (regardeless of the pathogen specificity), using a small sample volume.

When investigating cellular responses to viral infections, there is a need to quantitatively distinguish between dying cells, infected cells, and cells that are both infected and dying. Here, we present a protocol to simultaneously detect porcine reproductive and respiratory syndrome virus (PRRSV) infection and cell death using a flow cytometry double-staining approach. We describe steps for cell seeding, infection, and collection; staining dead cells; sample fixation; and cell permeabilization.

Stress negatively impacts university students, leading to adverse outcomes. While canine-assisted intervention (CAI) has been shown to reduce self-reported stress, no studies have investigated stress levels and associated biomarkers in dogs and students simultaneously. This study examined salivary cortisol, blood pressure, and pulse rate in 122 university students experiencing self-reported moderate to high stress before an encounter with a dog (T1), immediately before meeting a dog (T2), and after spending 15 minutes interacting with a dog (T3).

Mesh-reinforced reconstruction after abdominal wall desmoid tumor resection: a retrospective analysis of 16 cases.

Hernia

March 2025

Department of Hernia and Abdominal Wall Surgery, Peking University People's Hospital, No.11 Xizhimen South Street, Xicheng District, Beijing, 100044, China.

Purpose: Managing the defect after abdominal wall desmoid tumor resection is challenging due to the wide excision required. This report aims to review our institutional experience with mesh-reinforced reconstruction following desmoid tumor resection in the abdominal wall.

Methods: We retrospectively reviewed patients who underwent abdominal wall desmoid tumor resection with mesh-reinforced reconstruction between April 2014 and January 2019.
