A metal-free diastereo-/regioselective modular synthetic approach for the synthesis of highly constrained tetrahydroquinoline-fused tetracyclic heterocycles from easily available substrates has been developed. This two-step strategy utilizes an Ugi four-component reaction, followed by the intramolecular spirocarbocyclization and iodination reactions in a single operation. The transformation is mild and operationally simple, which provides architecturally complex polycyclic heterocycles with high diastereoselectivity. Furthermore, the preliminary cytotoxicity screening of selected compounds displayed promising anticancer activity against human cancer cell lines.

Delivery of therapies into skin is attractive for medical indications including vaccination and treatment of dermatoses but is highly constrained by the stratum corneum barrier. Microneedle (MN) patches have emerged as a promising technology to enable non-invasive, intuitive, and low-cost skin delivery. When combined with biodegradable polymer formulations, MN patches can further enable controlled-release drug delivery without injection.

Hardware implementation of reconfigurable and nonvolatile photoresponsivity is essential for advancing in-sensor computing for machine vision applications. However, existing reconfigurable photoresponsivity essentially depends on the photovoltaic effect of p-n junctions, which photoelectric efficiency is constrained by Shockley-Queisser limit and hinders the achievement of high-performance nonvolatile photoresponsivity. Here, we employ bulk photovoltaic effect of rhombohedral (3R) stacked/interlayer sliding tungsten disulfide (WS) to surpass this limit and realize highly reconfigurable, nonvolatile photoresponsivity with a retinomorphic photovoltaic device.

Pediatric solid tumors are a leading cause of childhood disease mortality. In this work, we examined germline structural variants (SVs) as risk factors for pediatric extracranial solid tumors using germline genome sequencing of 1765 affected children, their 943 unaffected parents, and 6665 adult controls. We discovered a sex-biased association between very large (>1 megabase) germline chromosomal abnormalities and increased risk of solid tumors in male children.

Photonic design is a process of mathematical optimization of a desired objective (beam formation, mode conversion, etc.) subject to the constraint of Maxwell's equations. Finding the optimal design is challenging: Generically, these problems are highly nonconvex and finding global optima is NP hard.

The NOD-like receptor protein 3 (NLRP3) inflammasome plays a crucial role in human acute and chronic liver diseases. However, the role and cell-specific contribution of NLRP3 in liver regeneration remains unclear. Here, we found that NLRP3 was highly activated during the early stage of liver regeneration via 70% partial hepatectomy (PHx) mice model and clinical data.
