Isoflavonoids are one of the most important groups of naturally occurring antioxidants. Their structural features are important for evaluating their antioxidative activity. In this work, density functional theory (DFT) methods were applied to investigate the influence of the C2=C3 double bond on the antioxidative activity of isoflavonoids based on three currently accepted radical scavenging mechanisms from the viewpoint of thermodynamics. The C2=C3 double bond can make the compounds more flat, which would extend the conjugated system in the molecule and make the isoflavonoids higher antioxidant activity. The C2=C3 double bond would not alter the strongest antioxidative hydroxyl group of the isoflavonoids. In the gas, benzene and CHCl phases, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids by lowering the bond dissociation enthalpies of the hydroxyl groups in the B ring that are the strongest antioxidative sites for the hydrogen atom transfer (HAT) mechanism. In polar phases, a similar result is obtained by weakening the proton affinity of 7-OH that is the strongest antioxidative hydroxyl group in the sequential proton loss electron transfer (SPLET), mechanism. Thus, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids irrespective of the studied phases.

The pathogenesis of diabetes is related to the amount of advanced glycation end products (AGEs) that are naturally generated from the attachment of glucose with tissue and circular proteins. Human serum albumin (HSA) is more susceptible to AGE occurrence than other circular proteins due to its sensitive sites and high abundance. Considering the location of hydroxyl groups in the structure of flavonoids, which play a major role in suppressing of AGEs generating pathways, the present study was conducted to compare the effect of the chemical peculiarities of five flavonoids: apigenin (AP), naringenin (NA), luteolin (LU), Quercetin (QU), and methylquercetin (MQ), in suppressing AGEs generated in the HSA/glucose system.

In the present work, the bond breaking/forming events along the intramolecular Diels-Alder (IMDA) reaction of (2,4,6)-2(allyloxy)cycloocta-2,4,6-trien-1-one have been revealed within bonding evolution theory (BET) at the density functional theory level, using the M05-2X functional with the cc-pVTZ basis set. Prior to achieving this task, the energy profiles and stationary points at the potential energy surface (PES) have been characterized. The analysis of the results finds that this rearrangement can proceed along three alternative reaction pathways (a-c).
