Deoxynivalenol (DON) is one of the most common fungal toxins that contaminate food grains and cereal-derived products. However, it is unknown whether DON stimulates IL-1β expression through the activation of the nuclear factor-κB (NF-κB) pathway and the ACS/NLRP3 inflammasome. In this study, we found that high concentrations of DON (above 800 nM) decreased relative cell viability; however, no significant population of apoptotic sub-G cells was observed. DON also upregulated expression from between 0.5 h and 6 h after treatment, and enhanced the nuclear localization of the NF-κB subunits, p50 and p65. NF-κB inhibitors, pyrrolidinedithiocarbamate and PS1145, significantly suppressed the DON-induced expression, which indicated that DON increased expression through the activation of NF-κB. In addition, marked secretion of IL-1β protein occurred in the presence of DON at 24 h, and a caspase-1 inhibitor suppressed DON-mediated IL-1β secretion, which suggested that caspase-1 induced the cleavage of pro-IL-1β to lead the secretion of its active form. Thus, components of the inflammasome, such as ASC and NLRP3, significantly increased by DON treatment; in addition, the knockdown of and markedly downregulated DON-induced IL-1β secretion, but not gene expression, which indicated that DON promoted IL-1β secretion through the ASC/NLRP3 inflammasome. Collectively, the data suggested that DON induced IL-1β expression in BV2 microglial cells through the activation of the NF-κB signaling pathway and the subsequent upregulation of the ASC/NLRP3 inflammasome. Therefore, DON may induce inflammatory diseases or disorders by activating IL-1β expression.

Article Synopsis
  • The study focuses on the detection of ASC aggregates, which are important for inflammasome assembly, particularly the NLRP3 inflammasome, after pyroptotic cell death.
  • Researchers developed a method using human monocytes, macrophages, and ASC reporter cells to identify ASC/NLRP3-positive events via flow cytometry.
  • Results showed increased ASC/NLRP3 specks in the sera of patients with inflammatory conditions, indicating that FACS is a reliable detection method with potential diagnostic uses in autoinflammatory diseases.

Background: Cerebral ischemia/reperfusion (I/R) injury is a serious vascular disease with extremely high mortality and disability rate. Bakuchiol (BAK) was found in leaves and seeds of Psoralea corylifolia Linn and has been shown to decrease inflammation and reduce oxidative stress, while the mechanism of BAK in ameliorating cerebral I/R injury remains unclear.

Methods: Middle cerebral artery occlusion reperfusion (MACO/R) was used to establish mouse model.

Article Synopsis
  • Adult-onset Still's disease (AOSD) is a complex autoinflammatory disorder with unclear causes, and this study aims to explore its genetic background and potential treatment targets.
  • Researchers analyzed 60 AOSD patients using advanced genetic techniques and blood profiling to uncover rare genetic variants and inflammation markers.
  • Results indicated a higher frequency of certain genetic variants and significant elevation of specific cytokines and immune system markers in AOSD patients, suggesting a complicated genetic landscape that could lead to new approaches in treatment.

PIWI-interacting RNAs (piRNAs) are involved in the regulation of hypertrophic cardiomyopathy, heart failure and myocardial methylation. However, their functions and the underlying molecular mechanisms in diabetic cardiomyopathy (DCM) have yet to be fully elucidated. In the present study, a pyroptosis-associated piRNA (piR112710) was identified that ameliorates cardiac remodeling through targeting the activation of inflammasomes and mitochondrial dysfunction that are mediated via the thioredoxin-interacting protein (Txnip)/NLRP3 signaling axis.

Objective: To investigate the mechanism of 2, 6-dimethoxy-1, 4-benzoquinone (DMQ), an active ingredients in fermented wheat germ extract, for inhibiting NLRP3 inflammasome activation and alleviating septic shock in mice.

Methods: Cultured murine bone marrow-derived macrophages (BMDM) stimulated with lipopolysaccharide (LPS) were treated with DMQ, followed by treatment with Nigericin, ATP, and MSU for activating the canonical NLRP3 inflammasome; the noncanonical NLRP3 inflammasome was activated by intracellular transfection of LPS, and AIM2 inflammasome was activated using Poly A: T.In human monocytic THP-1 cells, the effect of Nigericin on inflammasome activation products was examined using Western blotting and ELISA.
