Article Abstract

Grapevine trunk diseases (GTDs) are a major threat to the wine and grape industry. The aim of the study was to investigate the antifungal activity against , , and of ε-polylysine, chitosan oligomers, their conjugates, and culture filtrates, and their binary mixtures with chitosan oligomers. In vitro mycelial growth inhibition tests suggest that the efficacy of these treatments, in particular those based on ε-polylysine and ε-polylysine:chitosan oligomers 1:1 / conjugate, against the three Botryosphaeriaceae species would be comparable to or higher than that of conventional synthetic fungicides. In the case of ε-polylysine, EC values as low as 227, 26.9, and 22.5 µg·mL were obtained for , , and , respectively. Although the efficacy of the conjugate was slightly lower, with EC values of 507.5, 580.2, and 497.4 µg·mL, respectively, it may represent a more cost-effective option to the utilization of pure ε-polylysine. The proposed treatments may offer a viable and sustainable alternative for controlling GTDs.

Human chitinolytic enzymes trigger growing interest, not only because a wide range of diseases and allergic responses are linked to chitinous components of pathogens, including their interplay with human enzymes, but also due to the increasing use of chitosans in biomedical applications. Here, we present a detailed side-by-side analysis of the only two human chitinases, chitotriosidase and acidic mammalian chitinase, as well as human lysozyme. By analyzing the cleavage of well-characterized chitosan polymers and defined chitin and chitosan oligomers, we report mild processivity and a quantitative subsite preference typical for GH18 chitinases for chitotriosidase and acidic mammalian chitinase.

Chitooligosaccharides (CHOS) or chitosan oligosaccharides (COS) are oligomers mainly composed of d-glucosamine (GlcN) units and structured in a positively charged, basic, amino molecule obtained from the degradation of chitin/chitosan through physical, chemical, or enzymatic methods. CHOS display physicochemical properties attractive to applications from the food to the biomedical field, such as non-toxicity to humans, high water solubility, low viscosity, biocompatibility, and biodegradability. These properties also allow CHOS to exert important biological activities, for example, antioxidant, antimicrobial, anti-inflammatory, immunomodulatory, antitumor, and hypocholesterolemic ones, besides to exhibit applications in food systems, technological, and nutraceutical potential.
