Bayesian Computation through Cortical Latent Dynamics.

Statistical regularities in the environment create prior beliefs that we rely on to optimize our behavior when sensory information is uncertain. Bayesian theory formalizes how prior beliefs can be leveraged and has had a major impact on models of perception, sensorimotor function, and cognition. However, it is not known how recurrent interactions among neurons mediate Bayesian integration. By using a time-interval reproduction task in monkeys, we found that prior statistics warp neural representations in the frontal cortex, allowing the mapping of sensory inputs to motor outputs to incorporate prior statistics in accordance with Bayesian inference. Analysis of recurrent neural network models performing the task revealed that this warping was enabled by a low-dimensional curved manifold and allowed us to further probe the potential causal underpinnings of this computational strategy. These results uncover a simple and general principle whereby prior beliefs exert their influence on behavior by sculpting cortical latent dynamics.

Background: Calcineurin, a protein involved in functions such as synaptic plasticity and neuronal survival, plays an important role in the pathophysiology of Alzheimer's disease. This study, randomized, investigated the effects of FK506 (FK), a calcineurin inhibitor, on the behavioral, histological, and biochemical alterations observed in models of neurotoxicity induced by NMDA or Aβ and in a transgenic model for AD, in addition to the organotypic culture model stimulated with NMDA.

Methods: This study involved models for AD to experiment injecting NMDA or Aβ1-42 into the hippocampus of male C57Bl/6 mice aged 8-12 weeks to induce a neurotoxicity model, treating double-transgenic APP/PS1 mice, expressing both mouse/human APP and mutant human PS1, with chronic FK506 for AD, in which, to enable NMDA or Aβ1-42 microinjections, another experiment including a stereotaxic surgery was performed on the C57Bl/6 mice.

Background: Apolipoprotein E4 (E4) is the strongest genetic risk factor for sporadic Alzheimer's Disease (AD), and aging is the greatest overall risk factor for AD. Many cellular and molecular changes occur within the brain throughout aging, one of which being the increased bone morphogenetic protein 4 (BMP4) signaling. As APOE and BMPs are known to interact in non-neuronal organs, we hypothesized that enhanced BMP signaling in the brain may interact with APOE in a genotype-dependent manner to initiate or exacerbate neuropathological cascades relevant to AD.
