and proteins P40 and P75 belong to a large family of secreted cell wall proteins that contain a carboxy(C)-terminal CHAP or NlpC/P60 superfamily domains. In addition to their peptidoglycan hydrolases activity, proteins in this family are specific antigens of pathogens, frequently responsible of interactions with the host. GG and BL23 purified P40 and P75 proteins have antiapoptotic activity by inducing the EGF/Akt pathway. The aim of this work was to study the genetics, phylogeny and dissemination of this family of proteins in the genus as well as their characteristics and likely function. The scrutiny of their DNA encoding sequences revealed the presence of minisatellite DNA in the P75 encoding gene of strains (B) with intraspecific indels that gave raise to four different alleles (B1-4), which are exclusive of this species. Phylogenic analyses suggest that both proteins are present mainly in the and phylogenomic groups. A P40 ancestral gene was possibly present in the common ancestor of , and . P75 is also present in and groups, but its evolution is difficult to explain only by vertical transmission. Antibodies raised against the N-terminal regions of P40 and P75 improved their immunological detection in culture supernatants as they recognized almost exclusively proteins of strains, highlighting their structural similarity, that allowed to detect them in different fermented dairy products that contained probiotic strains. Purified P40 and P75 proteins showed no evident lytic activity but they complemented BL23 A and B defective mutants, respectively, thus proving that they actively participate in cell division.

Background: Atopic dermatitis (AD) is a chronic inflammatory skin disease resulting from the complex interplay of genetic and environmental factors, meriting exploration using temporally dynamic biomarkers. DNA methylation-based algorithms have been trained to accurately estimate biological age, and deviation of predicted age from true age (epigenetic age acceleration) has been implicated in several inflammatory diseases, including asthma.

Objective: We sought to determine the role of epigenetic and biological aging, telomere length, and epigenetically inferred abundance of 7 inflammatory biomarkers in AD.

Search for Influence Factors in Lactobacilli with Probiotic Properties Isolated from Traditional Kazakh Foods.

Probiotics Antimicrob Proteins

March 2024

Laboratory of Genetics and Biochemistry of Microorganisms, Republican Collection of Microorganisms, 13/1 Valikhanov Street, 010000, Astana, Republic of Kazakhstan.

The aim of this study was to investigate the factors influencing the probiotic potential of lactobacilli isolated from traditional Kazakh foods, focusing on the identification and characterisation of multifunctional proteins. The basis of the methodological approach in this scientific study is an empirical, experimental study of the factors of influence produced by lactobacilli, which were obtained from traditional Kazakh foods and have pronounced probiotic properties. In this scientific work, results were obtained indicating that the expressed factors of probiotic action can perform adhesive and signalling functions by analogy with homologous proteins of pathogenic and commensal/probiotic bacteria.

Bacterial extracellular vesicles and associated functional proteins in fermented dairy products with .

Front Microbiol

April 2023

Laboratory of Lactic Acid Bacteria and Probiotics, Department of Biotechnology, Instituto de Agroquímica y Tecnología de Alimentos (C.S.I.C.), Valencia, Spain.

Cells of all kingdoms produce extracellular vesicles (EVs); hence, they are present in most environments and body fluids. produces EVs that have attached biologically active proteins (P40 and P75). In this study, EV and functional proteins were found in five different commercial dairy-fermented products carrying .

Background: Muscle denervation from trauma and motor neuron disease causes disabling morbidities. A limiting step in functional recovery is the regeneration of neuromuscular junctions (NMJs) for reinnervation. Stem cells have the potential to promote these regenerative processes, but current approaches have limited success, and the optimal types of stem cells remain to be determined.
