In situ and in vivo visualization and analysis of functional, endogenous biomolecules in living systems have generated a pivotal impact in our comprehension of biology and medicine. An increasingly adopted approach involves the utilization of molecular vibrational spectroscopy, which delivers notable advantages such as label-free imaging, high spectral density, high sensitivity, and molecule specificity. Nonetheless, analyzing and processing the intricate, multi-dimensional imaging data to extract interpretable and actionable information poses a fundamental obstacle.

Objective: The diagnosis of early osteoarthritis when therapeutic interventions may be most effective at reversing cartilage degeneration presents a clinical challenge. We describe a Raman arthroscopic probe and spectral analysis that measures biomarkers reflective of the content of predominant cartilage ECM constituents-glycosaminoglycans (GAG), collagen, water-essential to cartilage function. We compare the capability of Raman-probe-derived biomarkers to predict functional properties of cartilage to quantitative MRI and histopathology assessments.

The acidity of historic paper, a property crucial for its preservation, is thought to mainly depend on the type of sizing. However, this research shows that during its degradation, paper acidity increases mainly due to the formation of non-volatile carboxylic acids, which accelerate acid-catalysed hydrolysis. Whether and how this accumulation depends on paper composition has not been studied systematically so far.

β-carbolines (βCs) represent a large family of bioactive alkaloids, including norharmane and normelinonine F, known for their diverse pharmacological activities. The effects of these alkaloids may depend, among other factors, on their delivery, accumulation in different subcellular compartments, and interactions with biomacromolecules such as serum albumins. In this study, we investigated the pH dependence of the interactions between bovine serum albumin (BSA) and four βCs (norharmane, normelinonine F, and their corresponding N(9)-methyl derivatives) using UV-vis and fluorescence spectroscopy, combined with multivariate analysis and molecular docking.
