The alpha helical CytolysinA family of pore forming toxins (α-PFT) contains single, two, and three component members. Structures of the single component Eschericia coli ClyA and the two component Yersinia enterolytica YaxAB show both undergo conformational changes from soluble to pore forms, and oligomerization to produce the active pore. Here we identify tripartite α-PFTs in pathogenic Gram negative bacteria, including Aeromonas hydrophila (AhlABC). We show that the AhlABC toxin requires all three components for maximal cell lysis. We present structures of pore components which describe a bi-fold hinge mechanism for soluble to pore transition in AhlB and a contrasting tetrameric assembly employed by soluble AhlC to hide their hydrophobic membrane associated residues. We propose a model of pore assembly where the AhlC tetramer dissociates, binds a single membrane leaflet, recruits AhlB promoting soluble to pore transition, prior to AhlA binding to form the active hydrophilic lined pore.

Pakistani lignite (PLC) was thermally dissolved at 300 °C using isopropanol (IPA) to obtain a soluble portion (SP) and insoluble portion (ISP). Proximate analysis, ultimate analysis, Fourier transform infrared spectrometry (FTIR), thermogravimetric analysis (TG-DTG) and pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS) results were compared to explore the influence of the thermal dissolution process on the pyrolysis for PLC and ISP. Results showed that the thermal dissolution process mainly dissolved some light components of low-rank coal, and more phenols, aldehydes, esters and ethers were found in the SP, indicating that low-carbon alcohols can break the ether bridge bond in coal and generate oxygen-containing organic compounds (OCOCs).

Introduction: This study aims to develop immediate release tablet formulations of lornoxicam (LRX) using hot melt extrusion (HME)-based fused deposition modeling (FDM) focusing on the adjustment of drug release by arranging infill densities and evaluating microcrystalline cellulose II (MCC II) as a disintegrating agent for HME-FDM purposes. LRX is a poorly soluble drug that exhibits pH-dependent solubility with a high thermal degradation temperature. These characteristics make it an ideal model drug for the HME-based FDM technique.

Essential oils (EOs) are natural, volatile substances derived from aromatic plants. They exhibit multiple pharmacological effects, including antibacterial, anticancer, anti-inflammatory, and antioxidant properties, with broad application prospects in health care, food, and agriculture. However, the instability of volatile components, which are susceptible to deterioration under light, heat, and oxygen exposure, as well as limited water solubility, have significantly impeded the development and application of EOs.

Compared to the laboratory preparation of biochar, there is less research on the adsorption of antibiotics by industrial production of biochar in water. In this study, three types of industrial production biochar (peanut shell biochar, sludge biochar, and perishable waste biochar) were selected, and their adsorption performance for tetracycline in composite-polluted water was systematically studied. The results indicated that the Freundlich equation could well fit the adsorption isotherms of the three types of biochar for tetracycline.
