We studied immunocytochemical localization of thyrotropin-releasing (TRH) in the bullfrog pats intermedia. Many TRH-like immunoreactive terminals containing immunoreactive granular vesicles 80-150 nm in diameter (116 nm mean diameter) were found in the pars intermedia. Some TRH-like immunoreactive terminals were in synaptic contact with the intermedia cells. These findings support the hypothesis that TRH directly activates the intermedia cells via synapses to secrete alpha-melanocyte-stimulating hormone.

Background: Rathke cleft cysts (RCCs) are benign sellar/suprasellar lesions that result from mucin-secreting vestigial remnants within the pars intermedia of the pituitary gland. When symptomatic, they can present with retro-orbital headaches, visual field defects, and/or pituitary dysfunction.
Observations: A 35-year-old female presented with subacute retro-orbital headache, right ptosis, and blurred vision.

Adult tissue stem cells of the anterior pituitary gland, CD9/SOX2-positive cells, are believed to exist in the marginal cell layer (MCL) bordering the residual lumen of the Rathke's pouch. These cells migrate from the intermediate lobe side of the MCL (IL-MCL) to the anterior lobe side of the MCL and may be involved in supplying hormone-producing cells. Previous studies reported that some SOX2-positive cells of the anterior lobe differentiate into skeletal muscle cells.

The recent discovery of nonvisual photoreceptors in various organs has raised expectations for uncovering their roles and underlying mechanisms. In this work, we identified a previously unrecognized hormone-releasing mechanism in the pituitary of the Japanese rice fish (medaka) induced by light. Ca imaging analysis revealed that melanotrophs, a type of pituitary endocrine cell that secretes melanocyte-stimulating hormone, robustly increase the concentration of intracellular Ca during short-wavelength light exposure.
