Increased Antibiotic Susceptibility of Gram-Positive Bacteria in Cerebrospinal Fluid Compared to Broth.

Antibiotics (Basel)

December 2024

Department of Geriatrics, Evangelisches Krankenhaus Göttingen-Weende, 37075 Göttingen, Germany.

In hospital- and community-acquired central nervous system infections, resistant Gram-positive bacteria are an increasing therapeutic challenge. The present approach does not attempt to identify rapidly bactericidal therapies for susceptible pathogens but aims to improve methods to find antibiotic regimens for multi-resistant pathogens that are effective in vivo in spite of reduced in vitro susceptibility in culture media. Antibiotic susceptibility was tested in cerebrospinal fluid (CSF) and Mueller-Hinton broth (, methicillin-resistant , ) or brain-heart infusion ().

Objectives: Sentinel surveillance for bacterial pneumonia (SSBP) allows the monitoring of immunopreventable diseases. The results of the SSBP carried out at HOMI, Fundación Hospital pediátrico de la Misericordia, are presented.

Methods: The daily active search for cases was carried out in accordance with the protocol of the Pan American Health Organization from January 2016 to December 2022.

Article Synopsis
  • * A total of 530 patients were reported, with the majority exhibiting pneumonia; the study highlighted high rates of ICU admissions (50.6%) and notable resistance to antibiotics among meningial and non-meningial isolates.
  • * The findings indicated a decrease in PCV10 serotypes but an increase in the prevalence of serotypes 19A and 6C, leading to Colombia's decision to switch to the PCV13 vaccine in July 202

In a previous work, we demonstrated that nasally administered 090104 beneficially modulated the respiratory innate immune response and improved the protection against Respiratory Syncytial Virus and in mice. In this work, we aimed to evaluate whether the immunomodulatory 090104 strain was able to enhance the resistance against the respiratory infection induced by hypermucoviscous carbapenemase-producing (KPC-2) strains belonging to the sequence type (ST) 25. The nasal treatment of mice with 090104 before the challenge with multiresistant ST25 strains significantly reduced lung bacterial cell counts and lung tissue damage.

Article Synopsis
  • The study focuses on the characterization of two endolysins, Skl and Pal, with Pal being particularly significant for its effectiveness against severe diseases.
  • Both endolysins are identified as cysteine-amidases and have a structural similarity to the enzyme papain, with unique active sites for breaking down bacterial components.
  • Skl is highlighted as a potent anti-pneumococcal agent, effective against resistant strains, but its initial lysis efficiency is limited, which suggests a complex interaction with bacterial targets.
