Capturing CO is highly valued in the field of organic synthesis, especially underdeveloped dual-CO conversion. In this study, we detail a novel reductive cyclization of 2-indolylanilines with dual CO as a difunctional reagent in the presence of PMHS [poly(methylhydrosiloxane)], delivering methyl-substituted quinoxalines. Furthermore, another chemoselective cyclization with 2-pyrrolylanilines is also realized by converting mono-CO.

BACKGROUND Programmed death 1 (PD-1) inhibitors have demonstrated limited effectiveness in patients with microsatellite instability-high (MSI-H) colorectal cancer (CRC). Recent studies suggest that their efficacy can be enhanced when combined with anti-angiogenic agents. CASE REPORT We present a case of a 25-year-old woman with CRC harboring a KRAS mutation and MSI-H status, along with initially unresectable liver metastases.

The study quantitatively analyzes and compares the pharmacokinetics (PK) of methylprednisolone (MPL) in humans upon administration of various dosage forms. The PK parameters and profiles of MPL in healthy subjects were collected from 22 literature sources. A minimal physiologically based pharmacokinetic (mPBPK) model consisting of blood and two tissue (lumped liver and kidney, remainder) compartments with nonlinear tissue partitioning was applied to describe MPL disposition.

The nuclear matrix, a proteinaceous gel composed of proteins and RNA, is an important nuclear structure that supports chromatin architecture, but its role in human pluripotent stem cells (hPSCs) has not been described. Here we show that by disrupting heterogeneous nuclear ribonucleoprotein U (HNRNPU) or the nuclear matrix protein, Matrin-3, primed hPSCs adopted features of the naive pluripotent state, including morphology and upregulation of naive-specific marker genes. We demonstrate that HNRNPU depletion leads to increased chromatin accessibility, reduced DNA contacts and increased nuclear size.

Photothermal therapy (PTT) utilizing cyanine dyes (Cy) and nitric oxide (NO) gas therapy via BNN6 have demonstrated significant potential in cancer treatment. However, the rapid clearance of these small molecules from the body limits their accumulation at tumor sites, thereby reducing therapeutic efficacy. To address this, we employed the acid-sensitive nanomaterial ZIF-8 as a carrier to encapsulate Cy and BNN6, creating functionalized BNN6-Cy@ZIF-8 Nanoparticles (B-C@Z NPs) for the targeted delivery and release of Cy and BNN6 at tumor sites.
