Copper-mediated radical cyclization of naphthalenyl iododifluoromethyl ketones with olefins was successfully developed to generate a series of unprecedented gem-difluorodihydrophenanthrenones, especially 2,2-difluoro-3,4-dihydrophenanthren-1(2H)-one derivatives. This strategy features the use of cheap copper powder and excellent regioselectivity and diastereoselectivity, thus providing a facile approach for application in drug discovery and development. Preliminary mechanistic studies indicate the involvement of difluorinated radical intermediates. Density functional theory (DFT) calculation was performed to provide further evidence for regioselectivity.

1,n-Dual Π systems including 1,4-diene derivatives have been widely used as the elegant radical receptors to promote cascade radical additions to give the highly functionalized polycyclic scaffolds. However, the tedious and complicated preparation of the former deters broad utilization and compromises the practical value. Herein, a straightforward preparation of 1,4-diene derivatives was developed from the easily accessible alkynes and γ,δ-unsaturated carboxylic acids via electrochemical oxidation cyclization and Hofmann elimination.

Spiromeroterpenoids from the Higher Plants.

Meroterpenoids are a distinctive class of natural products found in various organisms, including animals, plants, bacteria, algae, and particularly fungi. Among them, spiromeroterpenoids, which have a spiro-ring connecting a terpenoid and a non-terpenoid moiety, are markedly unique. Currently, only a limited number of plants from the families Myrtaceae, Hypericaceae, Annonaceae, Asteraceae, and Lauraceae are known to biosynthesize spiromeroterpenoids.

Oximes are widely used precursors in synthetic chemistry due to their broad availability and versatile chemical properties, in which N-O bond fragmentation represents a key reactivity mode. However, these transformations typically require the use of oxygen-protected oximes, and a general strategy to directly utilize free oximes remains challenging due to their vulnerability to side reaction pathways, rendering low tendency towards N-OH bond cleavage. Here we report a unified platform to achieve direct cyclization of unprotected oximes with enals, as well as other coupling partners through dual copper/organophotoredox catalysis under green light irradiation.

School of Chemistry and Materials Science, Hangzhou Institute for Advanced Study, University of Chinese Academy of Sciences, Hangzhou, China.

Synthetic organic chemists continually draw inspiration from biocatalytic processes to innovate synthetic methodologies beyond existing catalytic platforms. Within this context, although 1,2-amino migration represents a viable biochemical process, it remains underutilized within the synthetic organic chemistry community. Here we present a biomimetic 1,2-amino migration accomplished through the synergistic combination of biocatalytic mechanism and photoredox catalysis.

NIR-Responsive Double Closed-Ring Isomer of a Diarylethene Fused Dimer Synthesized by Stepwise Photochemical and Oxidative Cyclization Reaction.

The double closed-ring isomer of a diarylethene fused dimer was synthesized by a combination of photochemical and oxidative cyclization reaction. The double closed-ring isomer of a diarylethene fused dimer has fixed π-conjugation in a rigid planar framework so that this compound has been long-desired. However, with respect to the photocyclization of a diarylethene dimer with strong electronic interaction, the second-step cyclization is known to be suppressed due to fast energy transfer into the closed-ring isomer generated in the first-step photocyclization.
