In a recreational use of 3,4-methylenedioxymethamphetamine (MDMA, "Ecstasy"), some but not all users are stricken with a serious serotonin (-hydroxytryptamine; -HT) syndrome. This raises a question as to whether there exist subpopulations that are more susceptible to MDMA intoxication. The hypothesis was tested with hyperthyroid versus euthyroid rats by measuring changes in body-core temperature ( ) and -HT in the hypothalamus. In the euthyroid rats, injection of MDMA at a recreationally relevant dose had no serious effect on . In contrast, the same dose was sufficient to evoke life-threatening hyperthermia in hyperthyroid rats. Neurochemical studies revealed that there was greater -HT efflux in the hyperthyroid than the euthyroid rats. These effects were blocked by pretreatment with M100907, a -HT receptor antagonist. In summary, our data support the hypothesis that individuals with hyperthyroidism are more susceptible to having a serious serotonin syndrome following MDMA administration.

Design of Small Non-Peptidic Ligands That Alter Heteromerization between Cannabinoid CB and Serotonin 5HT Receptors.

Activation of cannabinoid CB receptors (CBR) by agonists induces analgesia but also induces cognitive impairment through the heteromer formed between CBR and the serotonin 5HT receptor (5HTR). This side effect poses a serious drawback in the therapeutic use of cannabis for pain alleviation. Peptides designed from the transmembrane helices of CBR, which are predicted to bind 5HTR and alter the stability of the CBR-5HTR heteromer, have been shown to avert CBR agonist-induced cognitive impairment while preserving analgesia.

Background: S-pindolol has metabolic effects of potential benefit in cancer cachexia: reduced catabolism through nonselective β-blockade; increased anabolism through partial β2 receptor agonism; and increased appetite and reduced fatigue through central 5-hydroxytryptamine/serotonin receptor activity. A Phase 2a clinical trial demonstrated that S-pindolol can reverse weight loss and improve fat-free mass in patients with cancer-related weight loss. A comparative phase I bioavailability study of S-pindolol and racemic pindolol was performed to support the development of S-pindolol in cancer cachexia.

Background: Premature ejaculation (PE) is one of the most common diseases in andrology and leads to serious male sexual dysfunction. Although several targeted oral drug therapies are used to treat PE, they often face challenges related to imprecise targeting and adverse effects. Acupuncture has shown potential in prolonging ejaculation time and improving couples' sexual quality of life.

Article Synopsis
  • - The study investigates the effectiveness of intradermal acupuncture (IA) as an alternative therapy for major depressive disorder (MDD) in a trial involving 120 participants, comparing it with selective serotonin reuptake inhibitors (SSRIs) and sham acupuncture.
  • - After 6 weeks of treatment, the group receiving SSRIs combined with active IA showed significantly greater improvement in depression scores than those on SSRIs alone or SSRIs with sham acupuncture, with no serious side effects reported.
  • - MRI analyses indicated that IA treatment improved functional connectivity in the brain regions associated with mood regulation, suggesting its potential role as an effective adjunct therapy for MDD.
