The detailed investigation of methylene blue (MB) dye with ovalbumin (OVA) was examined by multispectroscopic and computational techniques. The experimental results of emission spectral studies displayed that the quenching behaviour of OVA with MB dye is due to static quenching mechanism. Isothermal titration calorimetry experimental results suggested that the binding of MB dye became favoured with the aid of a favourable entropy contribution and negative enthalpy. The absorption and circular dichroism spectral experiments showed that the binding of MB dye prompted conformational modifications to the secondary structure of OVA protein. The computational studies have been used to predict the binding region and the stability of MB in OVA protein.Communicated by Ramaswamy H. Sarma.

A triple signal amplified electrochemical aptasensor for the detection of bisphenol A (BPA) was developed for the first time based on gold nanoparticles (AuNPs), hemin/G-quadruplex DNAzyme, and exonuclease I (Exo I) assisted amplification strategies. The BPA aptamer (Apt) hybridized with the capture probe (CP) was fixed on the gold electrode (GE) to form the double-stranded DNA (dsDNA) structure. When BPA was present, the Apt was detached from the GE surface by specific recognition between the BPA and Apt, forming BPA-Apt complexes in solution.

Per- and polyfluoroalkyl substances (PFAS) have raised significant concerns; however, their accurate detection in aqueous environments remains a major challenge. In this study, a host-guest molecularly imprinted polymer-based electrochemical sensor with enhanced antifouling properties were developed using β-cyclodextrin embedded with methylene blue (βCD-MB MIP). This sensor demonstrated sensitive and selective quantification of perfluorooctanoic acid (PFOA) in real water samples.
