Published: June 2019

The direct catalytic dehydrative amidation of β-hydroxycarboxylic acids with amines is described. A biphenyl-based diboronic acid anhydride with a B-O-B skeleton is shown to be an exceptionally effective catalyst for the reaction, providing β-hydroxycarboxylic amides in high to excellent yields with a low catalyst loading (minimum of 0.01 mol %, TON up to 7,500). This hydroxy-directed amidation shows excellent chemoselectivity and is applicable to gram-scale drug synthesis.

Boronic acids have been widely applied in various biological fields, particularly achieving significant practical progress in boronic acid-based glucose sensing. However, boronic acids exhibit nonspecific binding to other nucleophiles, and the inherent lability of boronic esters in biological systems limits their further applications. Herein, we developed a stimuli-responsive controllable caging strategy to achieve photoresponsive spatiotemporally and nitroreductase-responsive cancer cell-selective glucose sensing.

Single compounds displaying a wide range of luminescent colors are attractive optical materials for sensor applications. In this study, we present the beneficial combination of a cyclometalated iridium(III) complex scaffold and boronic acid units for designing stimuli-responsive luminescent materials with various emission colors. Five iridium(III) complexes bearing a diboronic acid ligand (bpyB2) were synthesized: Ir(C^N)bpyB2 (C^N = 2-phenylpyridine (1), 2-(2,4-difluorophenyl)pyridine (2), 2-(4-methoxyphenyl)pyridine (3), benzo[h]quinoline (4), 1-phenylisoquinoline (5)).

Article Synopsis
  • * It highlights the effectiveness of gold nanoparticles on Lewis acidic metal oxides, like α-FeO, in catalyzing the conversion of dialkyl ethers and diborons at around room temperature, yielding high amounts of alkylboronates.
  • * Mechanistic studies show that the reaction occurs at the interface between the nanoparticles and metal oxides, where strong Lewis acid sites are formed, illustrating a novel technology for sustainably synthesizing valuable organoboron compounds.

Liver 3D cell models are regularly employed as a screening platform for predicting the metabolic safety of drugs, by monitoring the physiological responses of the spheroids, through the measurement of relevant markers of normal liver physiology, notably glucose. Measuring glucose levels within the spheroids and their surroundings provides insight into the metabolic homeostasis of liver cells and may be employed as an indication of potential drug-induced toxicity. Several ortho-aminomethyl phenylboronic acid (PDBA) glucose sensors have been developed.
