Horseradish peroxidase (HRP) isoenzyme C1a is one of the most widely used enzymes for various analytical methods in bioscience research and medical fields. In these fields, real-time monitoring of HRP activity is highly desirable because the utility of HRP as a reporter enzyme would be expanded. In this study, we developed a simple assay system enabling real-time monitoring of HRP activity by using biolayer interferometry (BLI). The HRP activity was quantitatively detected on a BLI sensor chip by tracing a binding response of tyramide, a substrate of HRP, onto an immobilized protein. This system could be applied to analyses related to oxidase activity, as well as to the functional analysis of recombinant HRP.

In this work, a peroxidase-like (POD-like) nanozyme of Fe/P-NC was synthesized by doping phosphorus (P) and nitrogen (N) to manipulate iron (Fe) activity centers, which showed catalytic activity and kinetics comparable to those of natural HRP. Based on the efficient POD-like activity of the Fe/P-NC nanozyme and cascaded catalytic reactions with acetylcholinesterase (AChE), we constructed a colorimetric, affordable and sensitive sensing platform to detect organophosphorus pesticides (OPs). In the presence of AChE, the POD-like activity of the prepared Fe/P-NC was suppressed, which weakened the Fe/P-NC-catalyzed oxidation of TMB.

A Cr-doped VO nanobelt (Cr/VO) with remarkable peroxidase-like activity was synthesized and coupled with uricase to catalyze the cascade reaction for detection of uric acid. Notably, the affinity of Cr/VO for 3,3',5,5'-tetramethylbenzidine dihydrochloride hydrate (TMB) and hydrogen peroxide (HO) is tenfold and 20-fold higher, respectively, than that of horseradish peroxidase (HRP). The Cr/VO exhibits highly reactive and stable peroxidase activity at temperatures of 20-60 ℃.

Enzymatic cascade reactions are widely utilized in food security, environmental monitoring, and disease diagnostics, whereas their practical application was hindered due to their limited catalytic efficiency and intrinsic fragility to environmental influences. Herein, a compartmentalized dual-enzyme cascade nanoreactor was constructed in metal-organic frameworks (ZIF-8) by a shell-by-shell growth method. ZIF-8 provided a good microenvironment to maintain the activity of enzymes and protected them against harsh conditions.

Prussian blue nanoparticles (PBNPs) have been identified as a promising candidate for biomimetic peroxidase (POD)-like activity, specifically due to the metal centres (Fe/Fe) of Prussian blue (PB), which have the potential to function as catalytically active centres. The decoration of PBNPs with desired functional polymers (such as amino- or carboxylate-based) primarily facilitates the subsequent linkage of biomolecules to the nanoparticles for their use in biosensor applications. Thus, the elucidation of the catalytic POD mimicry of these systems is of significant scientific interest but has not been investigated in depth yet.

In 2023, a national gathering was convened to strategize implementation of the recommendations across policy, practice, and research settings. Discussions highlighted that meaningful engagement of women living with HIV (recommendation 1) is foundational to implementing the other recommendations.
