Background: Neurite degeneration is increasingly suspected to represent a causal feature of mild cognitive impairment (MCI) and Alzheimer's disease (AD). Therefore, sensitive and specific imaging biomarkers of neuronal degeneration are needed to elucidate the mechanisms underlying cognitive impairment in MCI and AD. However, the recently developed Neurite Orientation Dispersion and Density Imaging (NODDI) MRI technique, used to measure the neurite density index (NDI), has some limitations.

Background: Inexpensive, non-invasive tests may improve the identification of persons at increased risk for cognitive decline and dementia. We compared impairment in odor identification and global cognition with neuro-imaging biomarkers to predict cognitive decline and dementia in the population-based Mayo Clinic Study of Aging (MCSA).
Method: At the 2008 assessment, 647 participants who were ≥ 55 years old with at least one follow-up had the following procedures: modified Blessed Information-Memory-Concentration Test (BIMCT), 12-item Brief Smell Identification Test (BSIT), brain magnetic resonance imaging (MRI), and Positron Emission Tomography (PET) amyloid imaging with 11C-Pittsburgh compound B (11C-PiB).

Background: The locus coeruleus (LC) is one of the earliest regions accumulating tau pathology in Alzheimer's Disease (AD). As the disease progresses, tau in the LC has been linked to increasing cortical tau and amyloid-beta (Aβ) pathologies and cognitive decline. Previous animal research suggested that novelty-like phasic LC activity protects against AD-related cognitive decline.

Background: Myelin loss occurs as part of neurodegenerative and neuroinflammatory conditions. Myelin water fraction (MWF), a surrogate of myelin content, can be estimated using multicomponent relaxometry MRI. In this study we investigate the relationship between 7289 plasma proteins and MWF to derive a protein-based indicator of myelin content.
