Biomarkers for psychiatric disorders in children and adolescents are urgently needed. This cross-sectional pilot study investigated quantitative electroencephalogram (qEEG), a promising intermediate biomarker, in pediatric patients with major depressive disorder (MDD) compared with healthy controls (HCs). We hypothesized that youth with MDD would have increased coherence (connectivity) and absolute alpha power in the frontal cortex compared with HC. qEEG was obtained in adolescents aged 14-17 years with MDD ( = 25) and age- and gender-matched HCs ( = 14). The primary outcome was overall coherence on qEEG in the four frequency bands (alpha, beta, theta, and delta). Other outcomes included frontal-only coherence, overall and frontal-only qEEG power, and clinician-rated measures of anhedonia and anxiety. Average coherence in the theta band was significantly lower in MDD patients versus HCs, and also lower in frontal cortex among MDD patients. Seven node pairs were significantly different or trending toward significance between MDD and HC; all had lower coherence in MDD patients. Average frontal delta power was significantly higher in MDD versus HCs. Brain connectivity measured by qEEG differs significantly between adolescents with MDD and HCs. Compared with HCs, youth with MDD showed decreased connectivity, yet no differences in power in any frequency bands. In the frontal cortex, youth with MDD showed decreased resting connectivity in the alpha and theta frequency bands. Impaired development of a resting-state brain network (e.g., default mode network) in adolescents with MDD may represent an intermediate phenotype that can be assessed with qEEG.

Purpose: To elucidate the structural-functional connectivity (SC-FC) coupling in white matter (WM) tracts in patients with major depressive disorder (MDD).

Methods: A total of 178 individuals diagnosed with MDD and 173 healthy controls (HCs) were recruited for this study. The Euclidean distance was calculated to assess SC-FC coupling.

Selecting the optimal dose of psilocybin for treating Major Depressive Disorder (MDD) and Treatment-Resistant Depression (TRD) is crucial for clinical development and regulatory approval. This meta-analysis evaluates psilocybin's efficacy and safety in treating MDD to determine the optimal dose and timing for clinical trials. A systematic review and Dose-Response Network Meta-Analysis (NMA) of Randomized Placebo-Controlled Clinical Trials (RCTs) registered with PROSPERO was conducted.

Background: Cortical morphometry is an intermediate phenotype that is closely related to the genetics and onset of major depressive disorder (MDD), and cortical morphometric networks are considered more relevant to disease mechanisms than brain regions. We sought to investigate changes in cortical morphometric networks in MDD and their relationship with genetic risk in healthy controls.

Methods: We recruited healthy controls and patients with MDD of Han Chinese descent.

Background: Patients with Alzheimer's Disease (AD) frequently manifest comorbid neuropsychiatric symptoms (NPS) with depression and anxiety being most prevalent. Previously we identified shared genetic risk loci between AD and major depressive disorder (MDD). In another study, we constructed a polygenic risk score (PRS) based on MDD-GWAS data and demonstrated its performance in predicting depression onset in LOAD patients.

Background: Research on hemodynamic responses in the prefrontal cortex during working memory tasks has mainly been performed in cognitive neuroscience research. However, the specific stimuli contributing to working memory deficits in major depressive disorder (MDD) are currently lacking. This study aims to compare working memory performance and prefrontal cortex activation in individuals with MDD and healthy controls during working memory tasks involving textual, visual, and emotional stimuli.
