Several protein expression systems can be used to get enzymes in required quantities and study their functions. Incorporating a polyhistidine tag is a beneficial way of getting various enzymes such as FDHs for industrial applications. The NAD dependent formate dehydrogenase from (CtFDH) can be utilized for interconversion of formate to carbon dioxide coupled with the conversion of NAD to NADH. In this study, N-terminal His tagged CtFDH (N-CtFDH) and C-terminal His tagged CtFDH (C-CtFDH) was constructed to learn the effect of His tag location on the activity and kinetic parameters of the enzyme. The solubility of proteins was not affected by tag position, however, an interference on the N-terminal region caused a deterioration in specific activity and the kinetic ability of enzyme. The obtained results indicated that the C-terminus of the enzyme is an appropriate region for tag engineering. The C-CtFDH has an approximately three-fold larger specific activity and two-fold higher catalytic efficiency than N-CtFDH. The results suggest that insertion of a His-tag at the N-terminal or C-terminal end of CtFDH has different effects on the protein and the N-terminal fragment of the protein is crucial for the function of CtFDH.

Genomic selection using white clover multi-year-multi-site data showed predicted genetic gains through integrating among-half-sibling-family phenotypic selection and within-family genomic selection were up to 89% greater than half-sibling-family phenotypic selection alone. Genomic selection, an effective breeding tool used widely in plants and animals for improving low-heritability traits, has only recently been applied to forages. We explored the feasibility of implementing genomic selection in white clover (Trifolium repens L.

In this first QTL mapping study of embryo size in barley, novel and stable QTL were identified and candidate genes underlying a significant locus independent of kernel size were identified based on orthologous analysis and comparison of the whole-genome assemblies for both parental genotypes of the mapping population. Embryo, also known as germ, in cereal grains plays a crucial role in plant development. The embryo accounts for only a small portion of grain weight but it is rich in nutrients.

Early root traits, particularly total root length, are heritable and show positive genetic correlations with biomass yield in perennial ryegrass; incorporating them into breeding programs can enhance genetic gain. Perennial ryegrass (Lolium perenne L.) is an important forage grass widely used in pastures and lawns, valued for its high nutritive value and environmental benefits.

Sensitivity of Functional Arterial Spin Labelling in Detecting Cerebral Blood Flow Changes.

Department of Neurology, Wuhan Brain Hospital, General Hospital of Yangtze River Shipping, Wuhan, Hubei, China.

Arterial spin labelling (ASL) is a non-invasive magnetic resonance imaging (MRI) method. ASL techniques can quantitatively measure cerebral perfusion by fitting a kinetic model to the difference between labelled images (tag images) and ones which are acquired without labelling (control images). ASL functional MRI (fMRI) provides quantitative perfusion maps by using arterial water as an endogenous tracer instead of depending on vascular blood oxygenation level.

Given the increasing urban population and frenetic mobility, understanding how individuals perceive crowding at large-scale events is crucial for effective crowd management and safety. This study focuses on Tokyo Big Sight in Japan exhibitions to examine participants' perceptions of peak crowding times, locations, and local density, and compare them with the actual measurements. Our methodology integrated questionnaires with beacon tag data.
