Article Abstract

Since (APMV) was identified in 2003, several other giant viruses of amoebae have been isolated, highlighting the uniqueness of this group. In this context, the tupanviruses were recently isolated from extreme environments in Brazil, presenting virions with an outstanding tailed structure and genomes containing the most complete set of translation genes of the virosphere. Unlike other giant viruses of amoebae, tupanviruses present a broad host range, being able to replicate not only in sp. but also in other amoebae, such as , a widespread, free-living organism. Although the cycle in has been analyzed, there are no studies concerning the replication of tupanviruses in other host cells. Here, we present an in-depth microscopic study of the replication cycle of in . Our results reveal that can enter and generate new particles with similar morphology to when infecting cells. establishes a well-delimited electron-dense viral factory in , surrounded by lamellar structures, which appears different when compared with different cells. Moreover, viral morphogenesis occurs entirely in the host cytoplasm within the viral factory, from where complete particles, including the capsid and tail, are sprouted. Some of these particles have larger tails, which we named "supertupans." Finally, we observed the formation of defective particles, presenting abnormalities of the tail and/or capsid. Taken together, the data presented here contribute to a better understanding of the biology of tupanviruses in previously unexplored host cells.

Giant viruses are crucial for marine ecosystem dynamics because they regulate microeukaryotic community structure, accelerate carbon and nutrient cycles, and drive the evolution of their hosts through co-evolutionary processes. Previously reported long-term observations revealed that these viruses display seasonal fluctuations in abundance. However, the underlying genetic mechanisms driving such dynamics of these viruses remain largely unknown.

Carbohydrate-mediated interactions between chloroviruses and the immune system.

Commun Biol

December 2024

Department of Molecular Cell Biology and Immunology, Amsterdam UMC, Vrije Universiteit Amsterdam, Amsterdam, The Netherlands.

Understanding the molecular mechanisms which drive and modulate host-pathogen interactions are essential when designing effective therapeutic and diagnostic approaches aimed at controlling infectious diseases. Certain large and giant viruses have recently been discovered as components of the human virome, yet little is known about their interactions with the host immune system. We have dissected the role of viral N-linked glycans during the interaction between the glycoproteins from six chloroviruses (belonging to three chlorovirus classes: NC64A, SAG, and Osy viruses) and the representative carbohydrate-binding receptors of the innate immune system.

A hypothesis of nucleosome evolution considering mutational analysis.

Genes Genet Syst

December 2024

Division of Biochemistry, Faculty of Pharmaceutical Sciences, Tohoku Medical and Pharmaceutical University.

Nucleosomes are complexes of DNA and histone proteins that form the basis of eukaryotic chromatin. Eukaryotic histones are descended from Archaean homologs; however, how this occurred remains unclear. Our previous genetic analysis on the budding yeast nucleosome identified 26 histone residues conserved between S.

Virophage infection mode determines ecological and evolutionary changes in a host-virus-virophage system.

ISME J

January 2024

Aquatic Ecology and Evolution, Limnological Institute, University of Konstanz, Universitätsstraße 10, Konstanz/Egg 78464, Germany.

Giant viruses can control their eukaryotic host populations, shaping the ecology and evolution of aquatic microbial communities. Understanding the impact of the viruses' own parasites, the virophages, on the control of microbial communities remains a challenge. Most virophages have two modes of infection.

Unlabelled: Borgs are huge extrachromosomal elements of anaerobic methane-oxidizing archaea. They exist in exceedingly complex microbiomes, lack cultivated hosts and have few protein functional annotations, precluding their classification as plasmids, viruses or other. Here, we used structure prediction methods to investigate potential roles for ∼10,000 Borg proteins.
