Background: Kidney failure is a prevalent condition with tendency for familial clustering in up to 27% of the affected individuals. Living kidney donor (LKD) transplantation is the optimal treatment option; however, in Canada, more than 45% of LKDs are biologically related to their recipients which subjects recipients to worse graft survival and donors to higher future risk of kidney failure. Although not fully understood, this observation could be partially explained by genetic predisposition to kidney diseases.

Enhanced penstock structural models significantly advance hydropower engineering, yet their increasing complexity introduces challenges. As model interactions intensify, predictability and comprehensibility decrease, complicating the evaluation of model accuracy and alignment with operational performance metrics and safety standards. This issue is particularly pronounced in dynamic modeling, where knowledge gaps hinder straightforward validation via observational data.

Global adoption of wind energy continues to increase, while improving the efficiency of turbine settings requires reliable wind speed (WS) models. The latest models rely on artificial intelligence (AI) optimizations which constructs tests on a range of novel hybrid models to examine the reliability. Gradient Boosting (GB), Random Forest (RF), and Long Short-Term Memory (LSTM) are used in new combinations for data pre-processing.
View Article and Find Full Text PDFAcetyl CoA synthetases (ACS) have emerged as drug targets for the treatment of cancer, metabolic diseases as well as fungal and parasitic infections. Although a variety of small molecule ACS inhibitors have been discovered, the systematic optimization of these molecules has been slowed by a lack of structural information regarding their mechanism of inhibition. Through a chemical genetic-based, synthetic lethal screen of the human fungal pathogen Cryptococcus neoformans, we identified an isoxazole-based ACS inhibitor with antifungal activity and exquisite selectivity for the C.

Introduction: Digital trials are a promising strategy to increase the evidence base for common interventions and may convey considerable efficiency benefits in trial conduct. Although paediatric intensive care units (PICUs) are rich in routine electronic data, highly pragmatic digital trials in this field remain scarce. There are unmet evidence needs for optimal mechanical ventilation modes in paediatric intensive care.
