Background: Sipuleucel-T is an autologous cellular immunotherapy that is FDA approved for the treatment of asymptomatic or minimally symptomatic metastatic castrate-resistant prostate cancer (mCRPC). The IMPACT registry trial demonstrated a 4.1 month survival benefit, but not a consistent PSA response or improvement in progression-free survival. Based upon several factors, including this lack of objective treatment response, sipuleucel-T has been under-utilized in this patient population, despite current NCCN recommendations.

Methods: In order to explore if delayed treatment response occurs in a subset of patients, we performed a single institutional retrospective analysis of mCRPC patients treated with sipuleucel-T and ongoing ADT alone. Within that group, we then identified a subset of sipuleucel-T-treated men with long-term disease control and no additional interventions. To independently confirm this finding, we evaluated a total of 336 patients from 4 large urology group practices treated with sipuleucel-T between 2010 and 2014 and identified 44 patients who met the same criteria and demonstrated evidence of PSA stabilization post sipuleucel-T treatment.

Results: For this subgroup of patients, 79% (95% CI: 64.5%, 88.1%) survived 36 months with a median time to subsequent therapy of 17.8 months (95% CI 10.3, 25.3).

Conclusions: Although patient selection could account for some or all of these results, these data support the utilization of sipuleucel-T alone in select mCRPC patients that is associated with a delay in disease progression and a good overall prognosis.

MicroRNAs can be found intracellularly incorporated into extracellular vesicles (EV-miRNAs) or extracellularly as cell-free miRNAs (cf-miRNAs). This study aimed to compare the diagnostic and prognostic potential of four miRNAs with recognized roles in prostate cancer as cf-miRNAs and EV-miRNAs, obtained from liquid biopsies (LB). Total RNA was isolated from whole plasma and plasma EVs from 15 controls (CTR) and 30 patients (20 with localized prostate cancer (PCa), 10 with metastatic prostate cancer (mPCa)).

Achieving the optimal emergence profile: the role of soft tissue grafting and pontic site development.

Br Dent J

Specialty Registrar in Restorative Dentistry, Bristol Dental Hospital, Lower Maudlin Street, Bristol, BS1 2LY, UK.

Hard and soft tissue remodelling after tooth extraction may result in a concave profile at the subsequent edentulous ridge. This defect may result in a sub-optimal aesthetic transition zone between the soft tissue and the pontic, which may appear to sit on the ridge, rather than emanating from within the ridge, as would a natural tooth. To optimise aesthetics, pontic site augmentation (PSA) (increasing the volume at the pontic site) and pontic site development (PSD) (remodelling the tissue at the pontic site) may provide a solution.

Experimental optimization for synthesis of cerium-doped titanium dioxide nanoparticles by modified sol-gel process.

Department of Chemical Engineering, Collage of Engineering, Najran University, Najran, Saudi Arabia.

A systematic experimental optimization procedure was developed for the synthesis of cerium-doped titanium dioxide nanoparticles (CeTNPs) based on modified sol-gel process. The nanocomposite was prepared using titanium tetraisopropoxide (TTIP) as a catalyst precursor, while utilizing the non-ionic surfactant Triton X-114 in cyclohexane as a stabilizing agent. The synthesis process was optimized by identifying the main experimental factors that affect the properties of the nanoparticles, primarily the structural phase and particle size.

Background: Small molecule mimics offer the advantages of easy preparation, good thermodynamic stability, and reproducible catalytic activity. However, most of the reported organic artificial mimics face challenges including low catalytic activity, oxidative self-destruction, and auto-aggregation into inactive dimers. Therefore, novel organic mimics with high catalytic activity as well as good thermal and environmental stability are highly desirable.

Objective: The purpose of this study was to describe the developmental patterns of the posterior sloping angle (PSA) of the proximal femoral epiphyseal growth plate (PFEP) in a cohort of healthy Chinese children aged 5 to 14 years.

Methods: Pelvic frog-leg lateral radiographs of 400 healthy children (n=800 hips) were retrospectively analyzed. The cohort included 215 males and 185 females with a mean age of 9.
