Limited information exists about exposure to influenza A viruses (IAVs) in many wild waterbird species, including loons. We analyzed serum samples from breeding adult Pacific (), Red-throated (), and Yellow-billed () loons sampled at three locations along the coast of Alaska, US from 2008 to 2017 to gain a better understanding of the potential role loons play in IAV ecology. We screened loon sera for IAV antibodies using three tests-blocking enzyme-linked immunosorbent assay (bELISA), agar gel immunodiffusion (AGID), and hemagglutination inhibition (HI)-and examined patterns in seroprevalence among species and sampling locations. We found evidence of IAV infection in all loon species and at all breeding locations, although concordance was imperfect among serological tests. Diagnostic tests yielded seroprevalence estimates of 24% (42/172) with bELISA, 8% (5/60) with AGID, and 6% (4/70) with HI. The IAV subtypes to which loon sera reacted using HI were consistent with those detected in waterfowl and gulls at other locations in Alaska, suggesting that loons may be exposed to IAV maintained in sympatric waterbirds. Our study provided evidence that loons inhabiting Alaska were exposed to IAV. However, given imperfect concordance among serologic tests, and relatively low seroprevalence as compared to other avian taxa exposed to IAV in Alaska, they make poor IAV surveillance targets.

Immunogenic cell death (ICD) of tumor cells, which is characterized by releasing immunostimulatory "find me" and "eat me" signals, expressing proinflammatory cytokines and providing personalized and broad-spectrum tumor antigens draws increasing attention in developing a tumor vaccine. In this study, we aimed to investigate whether the influenza virus (IAV) is efficient enough to induce ICD in tumor cells and an extra modification of IAV components such as hemeagglutinin (HA) will be helpful for the ICD-induced cells to elicit robust antitumor effects; in addition, to evaluate whether the membrane-engineering polylactic coglycolic acid nanoparticles (PLGA NPs) simulating ICD immune stimulation mechanisms hold the potential to be a promising vaccine candidate, a mouse melanoma cell line (B16-F10 cell) was infected with IAV rescued by the reverse genetic system, and the prepared cells and membrane-modified PLGA NPs were used separately to immunize the melanoma-bearing mice. IAV-infected tumor cells exhibit dying status, releasing high mobility group box-1 (HMGB1) and adenosine triphosphate (ATP), and exposing calreticulin (CRT), IAV hemeagglutinin (HA), and tumor antigens like tyrosinase-related protein 2 (TRP2).

Maternal gut microbiota composition contributes to the status of the neonatal immune system and could influence the early life higher susceptibility to viral respiratory infections. Using a novel protocol of murine maternal probiotic supplementation, we report that perinatal exposure to () or () increases the influenza A/PR8 virus (IAV) clearance in neonates. Following either supplementation, type 1 conventional dendritic cells (cDC1) were amplified in the lymph nodes leading to an enhanced IAV antigen-experienced IFN-γ producing effector CD8 T cells in neonates and IAV-specific resident memory CD8 T cells in adulthood.
