Cotton, a natural fiber producing crop of huge importance, is often prone to attack of . Papain-like cysteine proteases (PLCPs) constitute a large family in plants and were proposed to involve in plant defense against pathogen attack in a number of studies. However, there is no detailed characterization of genes in cotton against infection of . In this study, we carried out a genome-wide analysis in cotton and identified seventy-eight PLCPs, which were divided into nine subfamilies based on their evolution phylogeny: RD21 (responsive to desiccation 21), CEP (cysteine endopeptidase), XCP (xylem cysteine peptidase), XBCP3 (xylem bark cysteine peptidase 3), THI, SAG12 (senescence-associated gene 12), RD19 (responsive to desiccation 19), ALP (aleurain-like protease) and CTB (cathepsin B-like). Genes in each subfamily exhibit a similar structure and motif composition. The expression patterns of these genes in different organs were examined, and subfamily RD21 was the most abundant in these families. Expression profiles under abiotic stress showed that thirty-five genes were induced by multiple stresses. Further transcriptome analysis showed that sixteen genes were up-regulated in response to in cotton. Among those, showed a higher transcription level than most other genes. Additionally, over-expression of led to enhanced resistance and RNAi lines were more susceptible to in cotton. Our results provide valuable information for future functional genomic studies of gene family in cotton.

However, there is no detailed characterization of PLCPs genes in poplar. In this study, a genome-wide analysis of the poplar PtrPLCPs family revealed 47 PtrPLCPs, which were classified into nine subfamilies according to their phylogeny: RD21, CEP, XCP, XBCP3, SAG12, RD19 (5), ALP, CTB, and the lost THI subgroups.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) papain-like cysteine protease (PLpro) represents one of only two essential cysteine proteases involved in the regulation of viral replication. It, therefore, qualifies as a promising therapeutic target for the development of antiviral agents. We identified a previously synthesized protease inhibitor, resulting from an earlier project, as a PLpro inhibitor and crafted a structure-activity relationship around the hit, leading to the more potent inhibitors ZHAWOC6941 (17h) and ZHAWOC25153 (17o) displaying IC values of 8 and 7 µM, respectively.
