Unlabelled: The combined effects of chronic ethanol ingestion and fasting (24-hr fast, except water, prior to delivery) were examined in term pregnant rats and their offspring. Rats fed liquid diet containing 5% (w/v) ethanol (EF) consumed fewer calories than those fed control diet and exhibited reduced weight gain relative to either ad libitum-fed (AF) or pair-fed (PF) controls. While the number of live fetuses at term was unaffected, fetal body and liver weights were reduced in EF litters. Blood glucose levels were significantly lower in EF fetuses although maternal glucose levels did not differ between the groups. Liver glycogen levels also were reduced in EF fetuses, without any change in plasma immunoreactive insulin or immunoreactive glucagon levels. Both active and total glycogen synthase and phosphorylase were significantly lower in livers of EF fetuses than in livers of control fetuses. After delivery, blood glucose and plasma immunoreactive insulin levels fell more slowly in EF neonates than in either control group, but EF neonates remained hypoglycemic at 4 hr postnatal, whereas glycemia in both control groups had recovered to normal. Plasma immunoreactive glucagon levels in EF were elevated during the first 2 hr following delivery relative to either AF or PF controls, and hepatic glycogen levels were reduced in EF neonates during the entire interval studied.
Conclusion: Fetal exposure to ethanol in utero and to a short maternal fasting prior to delivery results in fetal growth retardation, hypoglycemia, hypoinsulinemia, and liver glycogen depletion at term. Also, both glycogen synthase (active and total) and phosphorylase (active and total) were decreased as well.(ABSTRACT TRUNCATED AT 250 WORDS)

Sacubitril/valsartan, an angiotensin receptor neprilysin inhibitor (ARNI), is becoming more common in the treatment of heart failure and hypertension. Neprilysin is highly expressed in the renal tubules, and reports have shown increases in urinary C-peptide reactivity (CPR) levels after administration of ARNI. However, the effect of ARNI on serum CPR levels, a critical marker of insulin secretion in diabetes, remains underexplored.

This study aimed to elucidate the complexity of the humoral immune response in COVID-19 patients with varying disease trajectories using a SARS-CoV-2 whole proteome peptide microarray chip. The microarray, containing 5347 peptides spanning the entire SARS-CoV-2 proteome and key variants of concern, was used to analyze IgG responses in 10 severe-to-recovered, 9 nonsevere-to-severe cases, and 10 control case (5 pre-pandemic and 5 SARS-CoV-2-negative) plasma samples. We identified 1151 IgG-reactive peptides corresponding to 647 epitopes, with 207 peptides being cross-reactive across 124 epitopes.

Type 1 diabetes (T1D) is related to the autoimmune destruction of β-cells, leading to their almost complete absence in patients with longstanding T1D. However, endogenous insulin secretion persists in such patients as evidenced by the measurement of plasma C-peptide. Recently, a low level of insulin has been found in non-β islet cells of patients with longstanding T1D, indicating that other islet cell types may contribute to persistent insulin secretion.

The oncogenic and tumor suppressor roles of lnc-MAPKAPK5-AS1 in multiple cancers suggest its complexity in modulating cancer progression. The expression and promoter methylation level of lnc-MAPKAPK5-AS1 in hepatocellular carcinoma (HCC) was investigated through data mining from The Cancer Genome Atlas and Gene Expression Omnibus and its significance in prognosis and immunity was explored. lnc-MAPKAPK5-AS1 was co-expressed with its protein-coding gene MAPKAPK5 in HCC and exhibited upregulation in HCC tissues as a result of hypomethylation of its promoter region.
