We have developed a laboratory project in which students prepare liposomes, expose them to hyperosmotic and hypoosmotic solutions, and follow the resulting shrinking and swelling (respectively) with laser light scattering. Each light intensity transient can be fit to an exponential decline or rise, with the decay constant (k) and the amplitude (ΔV ) being indirectly related to the kinetics and thermodynamics (respectively) of transmembrane osmotic flux. Students vary the experimental system by changing the types and concentrations of osmolytes such as alcohols, amides, and salts. Students then compare how these changes alter the rate and extent of osmotic flux. This upper division biochemistry laboratory project is a challenging and rewarding one that exposes students to a biomolecule (lipid) and a spectroscopic technique (laser spectroscopy) that are not commonly used in the undergraduate laboratory setting. © 2019 International Union of Biochemistry and Molecular Biology, 47(3):239-246, 2019.

The high-sensitivity capabilities of laser-induced fluorescence (LIF) detection continuously promote the development of various labels with different fluorescence properties. However, this strategy also requires the adaptation of existing detection systems to suit the excitation and emission characteristics of novel fluorescent tags. Recently light-driven microdrones have been demonstrated, making use of plasmonic nanomotors based on directional resonant chiral light scattering. These nanomotors can be addressed individually, without requiring the tracking of a focused laser, leading to exceptional 2D maneuverability which renders microdrones a versatile robotic platform in aqueous environments. Here, we incorporate a light-operated manipulator, a plasmonic nano-tweezer, into the microdrone platform, rendering it a microrobot by enabling precise, all-optical transport and delivery of single nanoparticles suspended in solution.

The lipidome, encompassing the comprehensive lipid fingerprint of a biological system, includes thousands of unique isomeric and isobaric lipid species. Mass spectrometry (MS) is an effective technique for characterizing the lipidome, although the resolution of isomeric lipid species through MS typically requires specialized or modified equipment. In this study, we introduce a novel matrix derivatization technique that leverages the unique photoreactive properties of unsaturated lipids to reveal the double-bond location in conventional matrix-assisted laser desorption-ionization mass spectrometry (MALDI-MS) experiments.
