The discovery, structure-activity relationships, and optimization of a novel class of fatty acid synthase (FASN) inhibitors is reported. High throughput screening identified a series of substituted piperazines with structural features that enable interactions with many of the potency-driving regions of the FASN KR domain binding site. Derived from this series was FT113, a compound with potent biochemical and cellular activity, which translated into excellent activity in in vivo models.

Rapid temperature contrast hydrotherapy by water immersion has been utilized by athletes for effective sports recovery. However, its application at some training or competition venues is limited by high water consumption, bucky size, personal hygiene, and inconvenience. Here, a novel portable system equipped with highly effective, lightweight, and hygienic wearable fluidic fabric device is reported, that replaces direct water immersion.

Spatial transcriptomics (ST) technologies enable dissecting the tissue architecture in spatial context. To perceive the global contextual information of gene expression patterns in tissue, the spatial dependence of cells must be fully considered by integrating both local and non-local features by means of spatial-context-aware. However, the current ST integration algorithm ignores for ST dropouts, which impedes the spatial-aware of ST features, resulting in challenges in the accuracy and robustness of microenvironmental heterogeneity detecting, spatial domain clustering, and batch-effects correction.

Modern sequencing technologies allow for the addition of short-sequence tags, known as anchors, to both ends of a captured molecule. Anchors are useful in assembling the full-length sequence of a captured molecule as they can be used to accurately determine the endpoints. One representative of such anchor-enabled technology is LoopSeq Solo, a synthetic long read (SLR) sequencing protocol.

Here, we introduce a novel computational framework to model the dynamics of human behavioral choices by learning to align the temporal dynamics of a recurrent neural network (RNN) to human reaction times (RTs). We describe an approximation that allows us to constrain the number of time steps an RNN takes to solve a task with human RTs.

Introduction: To evaluate the enamel abrasion effects of soft, ultra-soft, and nano-bristle toothbrushes using atomic force microscopy (AFM) to guide toothbrush selection for optimal enamel preservation.
Methods: This in vitro study involved 45 extracted human teeth (central and lateral incisors), randomly assigned to three groups (n=15 each): Group I (nano-bristle), Group II (ultra-soft bristle), and Group III (soft bristle). Each specimen underwent 10,000 brushing cycles with a standardized 2 N force to simulate one year of brushing.
