Holographic projection for multispot structured illumination microscopy.

This paper describes the design and testing of a multispot structured illumination microscopy system using computer-generated holograms to create the required excitation patterns. Furthermore, it demonstrates the use of an adapted direct search algorithm for calculating the holograms that allows for imaging across an extended field of view. The system was tested on fixed targets and live cells yielding a two times resolution increase over conventional diffraction-limited imaging. LAY DESCRIPTION: We present the design and testing of a multispot structured illumination microscopy system using computer-generated holograms to create the required excitation patterns. It demonstrates the use of an adapted direct search algorithm for calculating the holograms that allows for imaging across an extended field of view. The system was tested on fixed targets and live cells yielding a two times resolution increase over conventional diffraction-limited imaging. The results here demonstrate that holography provides an efficient means of pattern projection for MSIM imaging. It provides a significant improvement in the efficiency of pattern projection and more importantly it allows for the testing of more diverse excitation patterns than possible with amplitude-only projection. For example, PSF engineering using phase modulation can be easily incorporated into the calculated holograms, potentially generating subdiffraction structures in the excitation pattern. The ability to incorporate PSF engineering into SIM opens up holographic MSIM as a potential method for further increasing resolution with little or no change to the imaging system.

Binding gene-wide single-stranded nucleic acids to surface-immobilized complementary probes is an important but challenging process for biophysical studies and diagnostic applications. The challenge comes from the conformational dynamics of the long chain that affects its accessibility and weakens its hybridization to the probes. We investigated the binding of bacteriophage genome M13mp18 on several different 20-mer probes immobilized on the surface of a multi-spot, label-free biosensor, and observed that only a few of them display strong binding capability with dissociation constant as low as 10 pM.

The optimal mode for ultrasonic welding (USW) of the "PEEK-ED (PEEK)-prepreg (PEI impregnated CF fabric)-ED (PEEK)-PEEK" lap joint was determined by artificial neural network (ANN) simulation, based on the sample of the experimental data expanded with the expert data set. Instability mechanisms of repelling peak solutions in a multi-variable activator-inhibitor system.

We study the linear stability properties of spatially localized single- and multi-peak states generated in a subcritical Turing bifurcation in the Meinhardt model of branching. In one spatial dimension, these states are organized in a foliated snaking structure owing to peak-peak repulsion but are shown to be all linearly unstable, with the number of unstable modes increasing with the number of peaks present. Despite this, in two spatial dimensions, direct numerical simulations reveal the presence of stable single- and multi-spot states whose properties depend on the repulsion from nearby spots as well as the shape of the domain and the boundary conditions imposed thereon.

A pulse burst optical system has been developed, able to alter an energetic, ultrafast 10 ps, 5 kHz output pulse train to 323 MHz intra-burst frequency at the fundamental 5 kHz repetition rate. An optical delay line consisting of a beam-splitting polariser cube, mirrors, and waveplates transforms a high-energy pulse into a pulse burst, circulating around the delay line. Interestingly, the reflected first pulse and subsequent pulses from the delay line have orthogonal linear polarisations.
