Higher-order conditioning phenomena, including context conditioning and blocking, occur when conditioning to one set of stimuli interacts with conditioning to a second set of stimuli to modulate the strength of the resultant memories. Here we analyze higher-order conditioning in the nematode worm Caenorhabditis elegans, demonstrating for the first time the presence of blocking in this animal, and dissociating it from context conditioning. We present an initial genetic dissection of these phenomena in a model benzaldehyde/NHCl aversive learning system, and suggest that blocking may involve an alteration of memory retrieval rather than storage. These findings offer a fundamentally different explanation for blocking than traditional explanations, and position C. elegans as a powerful model organism for the study of higher order conditioning.

Blast neurotrauma has been linked to impairments in higher-order cognitive functions, including memory, attention, and mood. Current literature is limited to a single overpressure exposure or repeated exposures at the same level of overpressure. The brain is a biological system comprising nerve cells and orchestrates its embodied agent's perception, behavior, and learning in dynamic environments. The free-energy principle (FEP) advocated by Karl Friston explicates the local, recurrent, and self-supervised cognitive dynamics of the brain's higher-order functions. In this study, we continue to refine the FEP through a physics-guided formulation; specifically, we apply our theory to synaptic learning by considering it an inference problem under the FEP and derive the governing equations, called Bayesian mechanics.

Animals can adapt their reward expectancy to changes in delays to reward availability. When temporal relations are altered, associative models of interval timing predict that the original time memory is lost due to the updating of the underlying associative weights, whereas the representational models render the preservation of the original time memory (as previously demonstrated in the extinction of conditioned fear). The current study presents the critical test of these theoretical accounts by training mice with two different intervals in a consecutive fashion (short → long or long → short) and then testing timing behaviors during extinction where neither temporal relation is in effect.
