Probiotics are the healthy living bacteria when administered in adequate amounts confers health benefits in the host. The main objective of present study was to screen the bacteria for potential probiotic characters and enzyme production. The probiotic characters like tolerance to low pH, bile salts, antibiotic sensitivity, hydrophobicity and auto-aggregation properties were evaluated. Among all isolates and sp G3_4_1TO2 showed maximum potential probiotic characters and produced amylase enzyme by observing the halo zone around the colonies with the diameter 0.9 mm and 1.23 mm. sp G3_4_1TO2 produced maximum amylase when compared with . The protein yield was 55.4% with the specific activity of 88.9 U/mg and obtained 40.8% purification fold. The molecular weight of amylase enzyme determined by SDS PAGE was 95,000 Da. From the present study it was considered that sp G3_4_1TO2 was a potential probiotic bacteria producing maximum amylase enzyme.

In this study, we developed a double-layer colon-targeted microcapsule. It used the Maillard product of gelatin-isomaltooligosaccharide (GI180) and zein-shellac complex (ZS) as bio-based materials, plant extracts (MPL) and Lactobacillus plantarum JJBYG12 (JJBYG12) were co-encapsulated, endowing them with strong resistance to harsh environments and precise intestinal adhesion and targeting ability. The research results indicated that ZS11 exhibits hydrogen bonding and electrostatic interactions.

Layer-by-Layer (LbL) self-assembly encapsulation is a promising technology for the protection and delivery of lactic acid bacteria. However, laboratory-scale encapsulation is often time-consuming, involves intensive protocols tailored for small-scale operations, requires substantial amounts of energy and water, and results in a low yield of encapsulated biomass. Scaling-up this process to a bench-bioreactor scale is not simply a matter of increasing culture volume as different key parameters (not particularly relevant at lab scale) become critical, including biomass production, the number of polymer layers, and the biomass-to-polymer mass ratio.
