Designing novel diffusion-weighted pulse sequences to probe tissue microstructure beyond the conventional Stejskal-Tanner family is currently of broad interest. One such technique, multidimensional diffusion MRI, has been recently proposed to afford model-free decomposition of diffusion signal kurtosis into terms originating from either ensemble variance of isotropic diffusivity or microscopic diffusion anisotropy. This ability rests on the assumption that diffusion can be described as a sum of multiple Gaussian compartments, but this is often not strictly fulfilled. The effects of nongaussian diffusion on single shot isotropic diffusion sequences were first considered in detail by de Swiet and Mitra in 1996. They showed theoretically that anisotropic compartments lead to anisotropic time dependence of the diffusion tensors, which causes the measured isotropic diffusivity to depend on gradient frame orientation. Here we show how such deviations from the multiple Gaussian compartments assumption conflates orientation dispersion with ensemble variance in isotropic diffusivity. Second, we consider additional contributions to the apparent variance in isotropic diffusivity arising due to intracompartmental kurtosis. These will likewise depend on gradient frame orientation. We illustrate the potential importance of these confounds with analytical expressions, numerical simulations in simple model geometries, and microimaging experiments in fixed spinal cord using isotropic diffusion encoding waveforms with 7.5 ms duration and 3000 mT/m maximum amplitude.

In biological systems such as cells, the macromolecules, which are anisotropic particles, diffuse in a crowded medium. In the present work, we have studied the diffusion of spheroidal particles diffusing between cylindrical obstacles by varying the density of the obstacles as well as the spheroidal particles. Analytical calculation of the free energy showed that the orientational vector of a single oblate particle will be aligned perpendicular, and a prolate particle will be aligned parallel to the symmetry axis of the cylindrical obstacles in equilibrium.

Bicelles, an artificial disk-shaped lipid bilayer, are commonly used for the structural and functional characterization of membrane-bound proteins in an environment similar to that in intracellular membranes. Because the dynamics of the lipids that constitute bicelles exert a significant impact on the structure and function of the inserted proteins, we investigated the mobility of lipid molecules in bicelles composed of DMPC (14:0 PC) and DHPC (06:0 PC) using solution NMR and MD calculations. C R relaxation experiments for the acyl groups demonstrated that increasing bicelle sizes limit the rotational diffusion of acyl chain H-C bonds in DMPC.

Dynamics of a single anisotropic particle under various resetting protocols.

We study analytically the dynamics of an anisotropic particle subjected to different stochastic resetting schemes in two dimensions. The Brownian motion of shape-asymmetric particles in two dimensions results in anisotropic diffusion at short times, while the late-time transport is isotropic due to rotational diffusion. We show that the presence of orientational resetting promotes the anisotropy to late times.

In this article, we investigate the dynamics of self-organised suspensions formed by rod-like fd virus colloids. Two methods have been employed for analysing fluorescence microscopy movies: single particle tracking (SPT) in direct space and differential dynamic microscopy (DDM) in reciprocal space. We perform a quantitative analysis on this anisotropic system with complex diffusion across different self-assembled states, ranging from dilute and semi-dilute liquids to nematic and smectic organisations.

Isotropic sampling of tensor-encoded diffusion MRI.

Magn Reson Med

Department of Clinical Medicine, Aarhus University, Aarhus, Denmark.

Purpose: The purpose of this study is to develop a method for selecting uniform wave vectors for double diffusion encoding (DDE) to improve the accuracy and reliability of diffusion measurements.

Methods: The method relies on identifying orthogonal wave vectors with rotations, and representing these rotations as points on a three-dimensional sphere in four dimensions using quaternions. This enables an electrostatic repulsion algorithm to achieve a uniform distribution of these points.
